Ni卟啉与ctDNA相互作用的研究

摘要 合成了水溶性阳离子meso-四(对-甲基吡啶基)卟啉(H2TM PyP)的镍卟啉(Ni(Ⅱ)TM PyP),并通过荧光光谱、荧光偏振和紫外可见吸收光谱的变化以及阴离子猝灭实验研究了Ni(Ⅱ) TM PyP与小牛胸腺DNA(ctDNA)的相互作用,测得二者的结合常数。根据实验结果,推断出Ni(Ⅱ) TMPyP与ctDNA的作用方式受到浓度的影响:Ni(Ⅱ) TMPyP与低浓度ctDNA仅发生外部结合;而Ni(Ⅱ) TMPyP与高浓度的ct DNA发生嵌插作用。 The nickel complex(Ni(Ⅱ) TMPyP) of water-soluble cationic meso-tetrakis(pmethylpyridyl) porphyrin(H2 TMPy P) was synthesized,and the interaction between Ni(Ⅱ) TMPyP and calf thymus DNA(ct DNA) was studied based on the changes of fluorescence spectra,fluorescence polarization UV-vis absorption and anion quenching study. The binding constants of Ni(Ⅱ) TMPyP to calf thymus DNA(ctDNA) was measured. The experimental results,showed that the binding mode of Ni(Ⅱ) TMPy P and ctDNA was affected by the concentration: Ni(Ⅱ) TMPyP only undergoes simple external binding with low concentration of ctDNA;while Ni(Ⅱ) TMPyP may intercalate into high concentration of ctDNA.
