摘要
为筛选适合海南产巴戟天多糖的提取方法,以得率为评价指标,在单因素实验基础上,通过正交实验优化传统水提、酶法、超声波辅助提取巴戟天多糖工艺;采用1,1-二苯基-2-三硝基苯肼法、超氧阴离子自由基、2,2-联氮-双-(3-乙基苯并噻唑啉-6-磺酸)二铵盐法和还原力法比较3种提取方法的抗氧化性。结果表明,传统水提法最佳工艺条件为温度70℃,料液比1:20(g/mL),浸提时间2.0 h;酶法提取巴戟天多糖的最佳工艺条件为纤维素酶用量1500 U/g,温度50℃,pH5.0,底物质量浓度50 g/L,时间1.5 h;超声波辅助提取法最佳工艺条件为超声功率300 W,温度70℃,料液比1:35(g/mL),浸提时间30 min。3种提取方法得到的巴戟天多糖均具有较强抗氧化性,对DPPH自由基清除作用较明显,其中酶法辅助提取的多糖清除DPPH自由基、O2-自由基、ABTS自由基能力最强。巴戟天多糖提取采用酶辅助较为适宜。
The aim was to select the suitable method for extracting polysaccharidess from Morinda officinalis How in Hainan, used extraction yield as evaluation index, the polysaccharidess was extracted by hot water extraction, compound enzymatic extraction and ultrasonic-assisted extraction. Antioxidant activities of polysaccharidess were evaluated by four different methods, 1,1-diphenyl-2-picrylhydrazyl(DPPH), superoxide anion(O2-), 2,2’-azinobis(3-ethylbenzothi azoline-6-sulfonic acid) ammonium salt(ABTS) radical scavenging assays and potassium ferricyanide reduction method. The results showed that the optimized extraction conditions of polysaccharides were, hot water extraction: extraction temperature70 ℃, material to liquid ratio 1:20(g/mL), time 2.0 h;compound enzymatic extraction additional:amount of cellulose 1500 U/g, extraction temperature 50 ℃, pH value 5.0, time 1.5 h and the substrate concentration 50 g/L;ultrasonic-assisted extraction: ultrasonic power 300 W, temperature 70 ℃, material to liquid ratio 1:35(g/mL), time 30 min. Meanwhile, The antioxidant activity assays showed that the three polysaccharides presented strong antioxidant activity. All three polysaccharides exhibited potent DPPH radical scavenging capacity. The polysaccharides with enzyme extraction had the strongest ability to scavenge DPPH free radical, O2-free radical and ABTS free radical. The extraction of polysaccharides from Morinda officinalis How were suitable for enzyme assistation.
