酶法糖基化修饰对玉米醇溶蛋白促酒精代谢活性的影响 被引量:1

Effect of enzymatic glycosylation on the alcohol-promoting activity of zein

摘要 以含有伯胺基团的壳寡糖为酰基受体,通过转谷氨酰胺酶催化的糖基化反应修饰玉米醇溶蛋白,用体外化学法和动物实验研究了糖基化修饰对玉米醇溶蛋白促酒精代谢活性的影响。实验结果表明,糖基化修饰可以改善玉米醇溶蛋白的促酒精代谢活性,且体外模拟胃肠消化不能破坏糖基化玉米醇溶蛋白的促酒精代谢活性。在蛋白质浓度为2 mg/mL时,糖基化玉米醇溶蛋白的乙醇脱氢酶激活率和羟基自由基清除率为11.83%和20.36%,分别比玉米醇溶蛋白高10.27%和10.57%。在糖基化玉米醇溶蛋白的灌胃剂量为2.0 mg/g bw时,小鼠血液中乙醇浓度为34.49mg/100 mL,与酒精模型组相比降低43.10%,且糖基化玉米醇溶蛋白的剂量与小鼠血醇含量的降低呈现明显的量效关系。 In the presented work, it’s exploited chitosan with containing primary amino groups as an acyl acceptor to modify zein by glycosylation reaction catalyzed by transglutaminase. Effect of enzymatic glycosylation on the alcohol-promoting activity of zein using chemical method in vitro and animal experiment was studied. The results showed that the alcohol-promoting activity of zein was improved by glycosylation and the activity could not be destroyed by in vitro gastrointestinal digestion. The alcohol dehydrogenase activation rate and hydroxyl radical scavenging rate of the glycosylated zein at2 mg/mL were 11.83% and 20.36%, a lot higher than that of zein which was 10.27% and 10.57%,respectively. The blood alcohol concentration in mice of the glycosylated zein group at 2.0 mg/g bw which was 34.49 mg/100 mL, which was decreased by 43.10% compared with the alcohol model group.Moreover, the decrease of blood alcohol concentration was found positively correlated with the dose of the glycosylated zein.
