西洋参血清指纹图谱及其抗疲劳谱效关系初步观察 被引量:11

摘要 目的观察灌胃给药西洋参后大鼠血清指纹图谱与抗疲劳间的谱效关系,以探讨西洋参抗疲劳的物质基础。方法利用UPLC-Q-TOF-MS建立给药后大鼠血清指纹图谱,并通过大鼠游泳时间、肝糖原含量、血尿素氮(BUN)含量和血清乳酸(LA)含量观察西洋参的抗疲劳作用,用偏最小二乘回归(PLSR)法对西洋参入血成分指纹峰与抗疲劳指标进行相关性分析。结果从西洋参血清指纹图谱中筛选出8个入血成分,通过PLSR分析发现人参皂苷Re、Rb1、Rb2与抗疲劳呈显著正相关性。结论人参皂苷Re、Rb1、Rb2为西洋参抗疲劳的潜在生理活性物质。 Objective To determine the spectrum-effect relationship between the serum fingerprints of Panacis Quinquefolii Radix and anti-fatigue effect after its ig administration to rats,and to elucidate its effective substances.Methods Ultra performance liquid chromatography with quadrupole and time-of-flight mass spectrometry(UPLC-Q-TOF-MS)was used for the fingerprint profiling by analyzing the serum samples obtained after the ig administration of Panacis Quinquefolii Radix to rats.The anti-fatigue effect of Panacis Quinquefolii Radix was observed by the time of weight-bearing swimming,content of hepatic glycogen,blood urea nitrogen(BUN)and blood lactic acid(LA)in the rats.The relationship between the absorbed fingerprints and pharmacodynamic parameters of Panacis Quinquefolii Radix was established by correlation analysis with partial least square regression(PLSR)method.Results Eight components were selected from the fingerprints of Panacis Quinquefolii Radix,among which ginsenoside Re,Rb1 and Rb2 were positively related to the pharmacodynamic data.Conclusion Ginsenoside Re,Rb1 and Rb2 are potential effective substances with antifatigue effects in Panacis Quinquefolii Radix.
