摘要
本研究在传统水提醇沉法基础上辅以酶消化法提取北五味子多糖,并用离子凝胶柱层析法对其进一步纯化,分离得到三种分子量的多糖组分(SCP-Ⅰ、SCP-Ⅱ和SCP-Ⅲ)。为分析多糖三种组分对鸡外周血淋巴细胞的作用,首先用高浓度(最高10mg/mL)的三种多糖组分作用于淋巴细胞,结果显示≤1mg/mL的多糖对细胞没有显著的毒性作用。以不同浓度的三种SCP组分(0、12.5、25.0、50.0、100.0、200.0、400.0、800.0、1600.0μg/mL)作用于淋巴细胞,发现SCP-Ⅲ对细胞因子IL-2、IL-6和IFN-γ的分泌具有显著的增强作用,但最佳作用浓度不同,SCP-Ⅰ和SCP-Ⅱ对这三种细胞因子的作用效果不显著。本研究为北五味子多糖的精制和应用奠定了理论基础。
In this study, the polysaccharide of Schisandra chinensis (SCP) was extracted by water extraction and alcohol precipitation method combined with enzyme preparation method. After purification by ion gel column chromatography, three polysaccharide components (SCP-Ⅰ, SCP-Ⅱ and SCP-Ⅲ) were obtained with different molecular weights. The three polysaccharide components with high concentration (up to 10 mg/mL) were applied to the peripheral blood lymphocytes of chickens to analyze their effects on lymphocytes. The results showed that ≤ 1 mg/mL of polysaccharide had no significant toxic effect on cells. Different concentrations of the three SCP components (0, 12.5, 25.0, 50.0, 100.0, 200.0, 400.0, 800.0, 1 600.0 μg/mL ) were applied to the lymphocytes, and the results showed that SCP-Ⅲ had significant enhancement effects on the secretion of cytokines IL-2, IL-6 and IFN-γ, but the optimal concentration was different. SCP-Ⅰand SCP-Ⅱ had no significant effect on these cytokines. This study laid a theoretical foundation for the purification and application of Schisandra chinensis polysaccharide.
