清毒伸筋汤对胶原诱导的关节炎大鼠血清VEGF含量的影响

Effect of Qingdu Shenjin Decoction on serum VEGF content in rats with collagen induced arthritis

目的观察清毒伸筋汤对胶原诱导的关节炎(CIA)大鼠血清血管内皮生长因子(VEGF)的影响,从细胞分子生物学角度探讨其治疗类风湿关节炎的机制。方法将实验大鼠分为正常组、模型组、甲氨蝶呤组,以及清毒伸筋汤高、中、低剂量组。正常组和模型组灌服生理盐水,其余各组予相应治疗药物灌服,连续治疗28d,评估各组大鼠关节炎指数(AI)评分、检测血清VEGF含量。结果清毒伸筋汤中、高剂量组在改善AI和下调VEGF方面优于模型组,差异有统计学意义(P<0.01)。结论清毒伸筋汤能下调CIA大鼠血清VEGF含量、降低AI,推测其能抑制VEGF的表达,从而抑制滑膜炎新生血管的形成,减轻关节骨质破坏和肿痛症状。

Objective To observe the effect of Qingdu Shenjin (QDSJ) Decoction on serum vascular endothelial growth factor (VEGF) in rats with collagen induced arthritis (CIA),and to explore the mechanism of QDSJ decoction in treating rheumatoid arthritis (RA) from the angle of cellular and molecular biology. Methods The experimental rats were divided into a normal control group,a model group,a methotrexate group,a high-dose QDSJ decoction group,a middle-dose QDSJ decoction group and a low-dose QDSJ decoction group. Rats in the normal control group and the model group received intragastric administration of normal saline. Rats in the rest groups were administered with corresponding drugs. All rats were treated consecutively for 28 days. The arthritis index (AI) score was evaluated and serum VEGF was measured for rats in each group. Results Compared with the model group,the high-dose QDSJ decoction group and the middle-dose QDSJ decoction group got better results in improving AI and down-regulation of VEGF,there were statistically significant differences ( P <0.01). Conclusion QDSJ decoction can down regulate serum VEGF and reduce AI in CIA rats. So it is speculated that QDSJ decoction can inhibit the expression of VEGF,thereby inhibit angiogenesis in synovitis and alleviate the joint bone destruction and the symptoms of joint swelling and pain.
