摘要
目的建立HPLC法测定中风合剂中黄芪甲苷和阿魏酸的含量.方法色谱条件为:色谱柱Agilent(250 mm×4.6 mm,5μm);流动相:乙腈(A)-0.05%磷酸水溶液(B)为22∶78;流速1.0 ml/min,检测波长285 nm,柱温30℃,进样量为10 μL,等度洗脱.结果黄芪甲苷和阿魏酸的线性回归方程分别为Y=33.051X+58.443,r=0.999 24;Y=27.763X+50.347,r=0.999 2;黄芪甲苷在20~200.0 μg/ml与峰面积积分值线性关系良好,阿魏酸在12.5~250.0μg/ml与峰面积积分值线性关系良好.测得中风合剂中黄芪甲苷和阿魏酸的平均含量分别为0.062 4mg/ml、0.132 6 mg/ml.结论本研究建立的HPLC方法可靠、准确、重现性好,为测定中风合剂中黄芪甲苷和阿魏酸的含量提供了参考.
Objective To establish a HPLC method for simultaneous determination of astragaloside and ferulic acid in Zhongfeng Mixture. Methods The chromatographic conditions were as follows: Agilent(250 mm×4.6 mm,5μm);mobile phase: acetonitrile(A)-0.05% phosphoric acid solution(B) =22∶78, flow rate 1.0 ml/min, detection wavelength 285 nm, column temperature 30℃, sample volume 10 μL, gradient elution. Results The linear regression equations of astragaloside A and ferulic acid were Y = 33.051 X + 58.443,r = 0.999 24;Y = 27.763 X + 50.347,r = 0.999 2.Astragaloside had a good linear relationship with the peak area integral value at 20 ~ 200.0 μg/ml, and ferulic acid at 12.5 ~ 250.0μg/ml. The average contents of astragaloside and ferulic acid were determined to be 0.0624 mg/ml and 0.132 6 mg/ml respectively. Conclusion The HPLC method established in this study is reliable, accurate and reproducible, which provides a reference for the determination of astragaloside and ferulic acid in Zhongfeng Mixture.
