摘要
目的:探讨成纤维细胞生长因子-21(FGF-21)在酒精性肝病中的保护作用及其机制。方法:建立酒精性肝病小鼠模型,比较两组小鼠生化指标和肝组织的病理学变化,Real-time PCR检测小鼠肝脏中过氧化物酶体增殖物激活受体γ辅激活酶因子1α(PGC-1α)的水平。结果:酒精喂养组小鼠肝脏中TG的含量、ALT和AST的含量、肝脏中PGC-1α的水平均显著高于对照饮食组(P<0.05)。结论:给酒精性肝病小鼠注射重组人FGF-21可以显著减轻肝脏内脂肪堆积,改善酒精对肝脏的损伤。
Objective: To investigate the protective effect and mechanism of fibroblast growth factor-21(FGF-21)in alcoholic liver disease. Methods: A mice model of alcoholic liver disease was established.The biochemical parameters and pathological changes of liver tissues were compared between the two groups.Real-time PCR was used to detect the level of peroxisome proliferator-activated receptor γ coactivator factor 1α(PGC-1α)in the liver of mice. Results: The levels of TG,ALT,AST and PGC-1α in the liver of the alcohol-fed mice were significantly higher than those in the control group( P <0.05). Conclusion: Injecting recombinant human FGF-21 into mice with alcoholic liver disease can significantly reduce the accumulation of fat in the liver and improve the damage of alcohol to the liver.
