摘要
目的:臭牡丹(CBS)在传统中医药中运用广泛,具有一定的抗菌性和提高人体免疫功能的功效,本文探讨CBS对地塞米松(DXMS)所致免疫抑制小鼠免疫功能的影响,为后续综合开发免疫性疾病新药提供参考。方法:通过不同浓度CBS提取物对致病大肠杆菌等6种细菌进行体外抑菌实验,根据外观和抑菌效果,选择合适供试品,用不同浓度CBS对免疫抑制小鼠进行腹腔注射,以云芝多糖(CVP)为阳性对照,检测腹腔免疫器官指数、巨噬细胞吞噬功能及T淋巴细胞亚群等免疫指标。结果:发现CBS水提物在125g/L时,对致病大肠杆菌等6种细菌均有理想的抑制作用;CBS提纯处理后在10~20g/L浓度范围内提高免疫器官指数及增强巨噬细胞吞噬功能,对剂量有依赖性,对免疫器官有明显保护作用,同时高浓度CBS使免疫抑制小鼠降低CD4^+T细胞数量、升高CD8^+T细胞数量、CD4^+/CD8^+T细胞数量明显降低至正常值,表明了高浓度CBS可使DXMS导致的细胞免疫功能正常化。结论:CBS可提高免疫抑制小鼠免疫器官及细胞的免疫功能,且效果优于CVP。
Objective: The Clerodendrum bungei steud(CBS) is widely used in traditional Chinese medicine, it has certain antimicrobial activity and improves the immune function of human body.This paper discusses the effect of CBS on the immune function of immunosuppressed mice induced by dexamethasone (DXMS),and provides a reference for the further comprehensive development of new drugs for immune diseases. Methods: Kinds of bacteria,such as Escherichia coli,were tested by different concentrations of peony extract in vitro.According to the appearance and bacteriostasis effect,the suitable test products were selected and the immunosuppressed mice were intraperitoneally injected with different concentrations of CBS.CVP was used as the positive control,the immune organ index of the peritoneal cavity was detected and the macrophage was swallowed.Immunological indexes such as phagocytic function and T lymphocyte subgroup. Results: It was found that the CBS aqueous extract had an ideal inhibitory effect on 6 kinds of bacteria such as Escherichia coli, in 125 g/L.The immune organ index and macrophage phagocytosis were enhanced in the concentration range of 10-20 g/L after CBS purification.The dosage was dependent on the dose,and the immune organs were obviously protected.At the same time,the high concentration of CBS made the immune suppression.In mice,CD8^+T cell amount and CD4^+/CD8^+ decreased to normal level by decreasing CD4^+T cell amount,indicating that high concentration of CBS can normalize cellular immune function induced by DXMS. Conclusion: CBS can improve immune organs and cellular immune function of immunosuppressed mice,and the effect is better than CVP.
