摘要 目的抗寄生虫环肽emodepside前体产生菌的鉴定及其次级代谢产物PF1022A结构分析。方法本实验室通过紫外诱变得到一株高产抗寄生虫环肽emodepside前体的突变株,采用形态观察、生理生化特征鉴定、ITS基因序列对比及系统发育学分析等相结合方法来确定该菌株的分离学地位,以及对该菌株经深层发酵,经LC-MS、1HNMR和13CNMR、DEPT90和DEPT135、1H-1HCOSY、HMQC、HMBC分析测定来确定产生次级代谢产物X的分子结构。结果与结论鉴定突变株CBR72-MCB-01呈现出座坚壳属(Rosellinia)菌株的形态学特征;经鉴定代谢产物结构与文献报道的抗寄生虫环肽PF1022A结构一致。 Objecive To identify a producing strain of an anthelmintic cyclodepsipeptide emodepside precursors and spectral analysis of its secondary metabolite PF1022A. Methods Polyphasic taxonomy based on morphology, physiological properties and ITS rRNA gene phylogenetic analysis were adopted to determine the taxonomic position of the strain. Based on the analysis of LC-MS, 1H NMR and 13C NMR, DEPT 90, and DEPT 135, 1H-1H COSY, HMQC, and HMBC spectra, the chemical structure of its secondary metabiolite, PF1022A was determined. Results and Conclusion A fungus, CBR72-MCB-01 with high yield by UV mutation, was identified as Rosellinia sp. by morphologic and culture characteristics. The chemical structure of its secondary metabolite PF1022A was known as anthelmintic cyclodepsipeptide produced by Rosellinia sp.
