基于高通量测序研究牛蒡子等五味中药合剂对皮肤浅部真菌病的作用机理 被引量:3

摘要 目的考察中药合剂治疗皮肤浅层真菌感染的作用机制。方法通过高通量测序技术分析以牛蒡子、金盏花、五加皮、凤仙草、地骨皮等五味中药所制的合剂,进行的相关抑菌实验的菌株细胞内全基因组的DNA变化情况。结果该中药合剂能明显的抑制3种供试菌株的活性,且具有一定的杀菌作用。结论通过高通量测序分析,结果发现该中药合剂能上调细胞结构破坏相关基因的表达,下调真菌细胞耐药性相关基因的表达,该中药合剂是通过降低细胞耐药性对细胞结构进行破坏进而达到抗菌效果的。 Objective:To investigate the mechanism of TCM mixture in the treatment of superficial skin fungal infection.Methods High-throughput sequencing technology was used to analyze the changes of whole-genome DNA in the bacterial strains of the relevant bacteriostasis experiment made by the mixture of five traditional Chinese medicines including arctium lengi, marigold, acanthopanax pentaphyllum, herba phoenixis and digupi.Results The mixture could obviously inhibit the activity of three tested strains and had certain bactericidal effect.Conclusion Through high-throughput sequencing analysis, it was found that the mixture could up-regulate the expression of genes related to cell structure destruction and down-regulate the expression of genes related to drug resistance in fungal cells. The mixture achieved the antibacterial effect by reducing cell resistance to damage cell structure.
