积雪草酸抑制去卵巢大鼠骨质疏松的作用

Exploring the effect of the asiatic acid on the osteoporosis of ovarian rats

目的探讨积雪草酸抑制去卵巢大鼠骨质疏松的作用。方法42只SPF级雌性SD大鼠按体质量参数随机分成6 组,分别为假手术组(Sham 组)、去卵巢组(OVX 组)、低\中\高剂量积雪草酸组(L\M\H-AA组)、雌二醇组(E2组)。Sham组切除双侧卵巢附近脂肪组织,其余5组则切除双侧卵巢组织,Sham组、OVX组每天灌胃生理盐水;L\M\H-AA组每天灌胃相应浓度的积雪草酸;E2组则每天灌胃雌二醇,实验周期为12周,每周称重一次,实验结束后,大鼠心脏取血处死,取出股骨进行指标检测。结果与Sham组相比,OVX组大鼠骨密度(BMD)、骨体积分数(BV/TV)、骨小梁数量(Tb.N)、骨小梁厚度(Tb.Th)、碱性磷酸酶(ALP)降低,低密度脂蛋白(LDL)、结构模型指数(SMI)、骨小梁分离度(Tb.Sp)、DKK1蛋白(DKK1)升高,差异具有统计学意义(P<0.05);与OVX组比较,L-AA组大鼠LDL、总胆固醇(TC)升高,差异具有统计学意义(P<0.05),M-AA组大鼠BMD、BV/TV、Tb.N、Tb.Th、ALP降低,甘油三酯(TG)、SMI升高,差异具有统计学意义(P<0.05),H-AA组大鼠BMD、Tb.N降低,LDL、TG升高,差异具有统计学意义(P<0.05),E2组大鼠BMD、Tb.Th、ALP降低,TC、TG、SMI、DKK1升高,差异具有统计学意义(P<0.05)。结论积雪草酸具有抑制去卵巢大鼠骨质疏松的作用。

Objective Study the effect of Asiatic acid on the osteoporosis of ovarian rats.Method 42 female rats were randomly divided into 6 groups according to the weight parameters--Sham group,OVX group,L/M/H-AA group,and E2 group.In the Sham group the fat near bilateral ovarian tissues was excised,while the bilateral ovarian tissues were excised in the other groups.For 12 weeks,the Sham group and OVX group were administered saline;the L/M/H-AA group were administered the corresponding concentration of Asiatic acid,and the E2 group was administered with Rocaltrol.Rats were weighed once a week and sacrificed after 12 weeks in order to take out femur and blood in the heart.Result Compared with the Sham group,in the OVX group,the index of BV/TV,bone density(BMD),Tb.N,Tb.Th,ALP,low density lipoprotein(LDL),SMI,and Tb.Sp decreased and Dickkopf1 protein(DKK1)increased with statistical difference(P < 0.05).Compared with the OVX group,in the L-AA group,LDL and TC increased with statistical difference(P < 0.05);in the M-AA group,BMD,BV/TV,Tb.N,Tb.Th,ALP decreased while TG and SMI increased with statistical difference(P < 0.05);in the H-AA group,BMD and Tb.N decreased while LDL and TG increased with statistical difference;in the E2 group,Tb.Th,BMD and ALP decreased while TC,TG,SMI and DKK1 increased with statistical difference(P < 0.05).Conclusion The asiatic acid had the effect of resisting the osteoporosis of the ovarian rats.
