摘要
目的:探讨肺癌组织中DNA聚合酶ζ(DNA polymeraseζ,polζ)的表达及其临床意义。方法:应用免疫组化技术检测108例肺癌组织及配对癌旁组织中polζ蛋白的表达,分析其与肺癌临床病理学特征的关系,并探讨其预后价值。结果:polζ在肺癌组织中,高表达率为49.1%(53/108),在癌旁组织中,高表达率为29.6%(32/108),两组之间行χ~2检验,差异有统计学意义(χ~2=8.555,P=0.003);polζ在癌组织不同TNM分期之间差异有统计学意义(χ~2=8.465,P=0.004);在不同性别、年龄、病理类型、病理分级组间未见明显差异,差异无统计学意义(P>0.05);polζ低表达组的生存明显优于高表达组,两组比较差异有统计学意义(χ~2=1.824,P=0.036)。COX多因素分析polζ被进一步剔除(P>0.05),TNM分期是影响预后的独立判定因素(P<0.05)。结论:polζ在肺癌组织中存在过表达,且与分期和肿瘤的预后相关,但不是影响预后的独立因子。
Objective To investigate the expression of DNA polymerase ζ in lung cancer and its relationship withprognosis. Method A total of 108 patients with lung cancer and adjacent tissues were collected.Immunohistochemistry (IHC) was used to analyze pol ζ in cancer and adjacent tissues.Chi-square test for the relationship among different groups of clinicopathological factors, and the OS of the different pol ζ expression group were analyzed by Log-rank test. Results Pol ζ was mainly stained in cytoplasm, the high expression was 49.1%(53/108) in lung cancer, while 29.6%(32/108) in adjacent tissues.There was a statistically significant difference (χ 2 =8.555, P =0.003).According to the expression of pol ζ, there were significant differences between different TNM stages(χ 2 =8.465, P =0.004), at different ages, sex, pathological types, pathological grades groups, no significant difference was observed between the two groups( P >0.05).Log-rank test showed that the OS of the low expression group were better than those of the high expression group(χ 2 =1.824, P =0.036).COX multivariate analysis pol ζ was further excluded, TNM staging was an independent determinant of prognosis. Conclusion Pol ζ is highly expressed in lung cancer and is associated with prognosis.It is a potential therapeutic target for lung cancer.
