摘要
【目的】通过CRISPR/Cas9基因组编辑技术遗传改良优质粳稻秀水134来创制糯稻新材料。【方法】构建靶向水稻直链淀粉合成主效基因Wx的CRISPR/Cas9表达载体,并通过农杆菌介导秀水134的遗传转化。【结果】共获得了45个转基因株系,其中41个株系发生靶定位置的碱基修改,突变率达到91.1%。通过表型考察和直链淀粉含量分析获得了糯稻新材料。【结论】该方法可以创制直链淀粉明显降低的糯稻新材料,为糯稻育种研究提供重要的材料基础。
【Objective】This study intends to genetically improve the quality of japonica rice Xiushui134 by the CRISPR/Cas9 genomic editing technology to create new glutinous rice materials.【Method】 A CRISPR/Cas9 vector targeted the Waxy gene, then transformed into the calli of Xiushui134 mediated with agrobacterium.【Result】 45 transgenic lines were obtained, among them, 41 lines were modified at the target site, and the mutation ratio reached 91.1%.New glutinous rice materials were obtained by phenotypic investigation and amylose content analysis.【Conclusion】 It was indicated that this method could obtain new glutinous rice materials with reduced amylose content significantly, which provides an important material basis for glutinous rice breeding research.
