17β-雌二醇促进大鼠坐骨神经横断性损伤修复 被引量:1

17β-estradiol promotes the repair of sciatic nerve transection injury in rats

摘要 目的:研究17β-雌二醇对大鼠RSC96雪旺细胞活性的影响及对SD大鼠坐骨神经横断性损伤的修复作用。方法:配制不同浓度的17β-雌二醇溶液,用CCK-8试剂盒检测雪旺细胞的增殖能力,筛选17β-雌二醇作用的最适浓度;将大鼠雪旺细胞分为对照组(常规培养)和17β-雌二醇组(100 nmol/L),用蛋白印迹法检测细胞雌激素受体的表达;用Transwell实验和划痕实验检测细胞的迁移能力;用qRT-PCR检测脑源性神经营养因子(brain derived neurotrophic factor,BDNF)、睫状神经营养因子(ciliary neurotrophic factor,CNTF)、胶质细胞源性神经营养因子(glialcellline-derived neurotrophic factor,GDNF)、神经生长因子(nerve growth factor,NGF)和血管内皮生长因子(vascular endothelial growth factor,VEGF)等mRNA的表达;雄性SD大鼠经腹腔注射一定浓度的17β-雌二醇溶液,观察其对坐骨神经横断性损伤的修复作用。结果:100 nmol/L 17β-雌二醇溶液细胞增殖能力明显强于其他浓度(P <0. 05);与对照组相比,100 nmol/L 17β-雌二醇溶液细胞增殖能力明显增强,雌激素受体表达明显增加,细胞迁移能力显著增强,BDNF、CNTF、GDNF、NGF等mRNA表达明显增加(P均<0. 05)。同时,17β-雌二醇促进SD大鼠坐骨神经损伤后运动和感觉功能的恢复,延缓腓肠肌的萎缩。结论:17β-雌二醇可能通过与雪旺细胞上雌激素受体结合来发挥一系列的生物学效应,促进外周神经损伤后运动和感觉功能的恢复。 Objective: To study the effect of 17β-estradiol on the activity of Schwann cells in rats and the repair effect of 17β-estradiol on transection injury of rat sciatica. Methods: Different concentrations of17β-estradiol solution was prepared,and the proliferation of Schwann cells was detected by CCK-8 kit to select the optimal concentration;the expression of estrogen receptor on Schwann cells was determined by Western blotting. Schwann cells were divided into two groups,control group( routinely cultured) and17β-estradiol group( 100 nmol/L);and the migration ability of Schwann cells was detected by Transwell test and scratch test. The mRNA expression of brain-derived neurotrophic factor( BDNF),ciliary neurotrophic factor( CNTF),glial cell-derived neurotrophic factor( GDNF),nerve growth factor( NGF) and vascular endothelial growth factor( VEGF) were quantitatively detected by qRT-PCR. A certain concentration of17β-estradiol was administered by intraperitioneal injection to observe the repair of transection injury of sciatica nerve in male SD rats. Results: Compared with the control group,17β-estradiol of 100 nmol/L increased the proliferation of Schwann cells,promoted the expression of estrogen receptors on Schwann cells,and increased the migration ability of Schwann cells,the neurotrophic factor BDNF,CNTF,GDNF and NGF mRNA levels were increased( all P < 0. 05). 17β-estradiol could promote the recovery of motor and sensory function after sciatic nerve injury in rats,and delay the atrophy of the gastrocnemius( all P <0. 05). Conclusion: 17β-estradiol may exert a series of biological effects by binding to estrogen receptors on Schwann cells,promoting the recovery of motor and sensory functions after peripheral nerve injury.
