Expression and clinical significance of long non-coding RNA LINC00858 in gastric cancer

目的检测长链非编码RNALINC00858在人胃癌及其癌旁组织中的表达情况,分析LINC00858表达量变化与人胃癌发生、发展的关系。方法利用在线统计软件分析TCGA数据库中LINC00858在人胃癌中的表达情况及其表达量变化与胃癌患者总生存期、无病生存期及患者临床分期间的关系。用实时荧光定量PCR法检测LINC00858在胃癌及其配对癌旁正常组织中的表达情况。统计学方法分析LINC00858表达量与胃癌患者年龄、性别及临床分期的关系。结果LINC00858在三种肾癌和骨髓癌组织中的表达量有所下降,在胃癌及其他类型的肿瘤组织中呈上升趋势;胃癌患者肿瘤组织中LINC00858表达量越高患者的临床分期越晚。结论LINC00858表达量的升高与胃癌的发生、发展相关,LINC00858表达量的检测及调控可能有助于临床胃癌患者病情的监测及治疗。

Objective To investigate the relative expression change of long non-coding RNA LINC00858 in gastric cancer compared with that in normal adjacent tissues,and further analyze the relationship between LINC00858 and gastric cancer development. Methods Relationships between LINC00858 expression and gastric cancer patients’ overall survival,disease-free survival and clinical stage were statistically analyzed by The Cancer Genome Atlas (TCGA) data.Expressions of LINC00858 in 20 gastric cancer tissues and paired normal adjacent tissues were tested by real-time PCR.Relationship of LINC00858 expression with patients’ age,sex,clinical stage were analyzed by Graphpad Prism 7.0. Results Expressions of LINC00858 were decreased in three kinds of renal cell carcinoma and bone cancer tissues,while increased in gastric cancer and other types of tumor tissues.In gastric cancer patients,expression of LINC00858 was higher in tumor tissues than that in normal adjacent tissues,and the more lately clinical staged,the higher LINC00858 expressed. Conclusion Increased LINC00858 expression is related to the occurrence and development of gastric cancer.Increased LINC00858 expression may be used in gastric cancer diagnose and clinical monitor.
