葛根素对CVB3病毒致心肌炎模型miRNA表达的影响

Effect of Puerarin on CVB3 miRNA Expression of Viral Myocarditis Model

目的:研究葛根素对CVB3病毒所致小白鼠病毒性心肌炎模型中miRNA表达的影响。方法:选取昆明种小白鼠60只,随机分为空白对照组、葛根素低剂量组、葛根素中剂量组、葛根素高剂量组、阳性药物(黄芪注射液)对照组和模型对照组,每组10只。除空白对照组外,其余50只小白鼠腹腔注射CVB3标准病毒株建立小白鼠病毒性心肌炎模型。8d后,腹主动脉取血,测定血清中SOD酶活性和MDA含量。取心脏,沿冠状面纵切,前部分制组织匀浆,测定清液中SOD酶活性和MDA含量。后部分心房部分测定组织中CVB3 miRNA的表达;心室部分制作病理切片,观察结构。结果:葛根素三个剂量组对病毒性心肌炎模型血清和心肌组织匀浆中的SOD酶活性和MDA含量有不同程度的改善。同时,对CVB3 miRNA的表达都有一定的抑制作用。葛根素在影响模型血清中的SOD酶活性和MDA含量和CVB3 miRNA的表达过程中均呈现剂量依赖性,即高剂量组优于中剂量组优于低剂量组。结论:葛根素对CVB3所致小白鼠病毒性心肌炎模型中CVB3 miRNA的表达有较好的抑制作用。

Objective:Puerarin on mice viral myocarditis caused by CVB3 model affect CVB3 miRNA expression. Methods:Choose 60 mice,Randomly divided into control group,puerarin low dose group,middle dose group puerarin,puerarin high-dose group,positive drug(Huangqi) in the control group and model group,which group have 10 mice. In addition to the control group,the remaining 50 mice were injected CVB3 standard strains of mice with viral myocarditis model establishment.8 days later,Take heart,determination of tissue CVB3 miRNA expression levels measured in serum SOD activity and MDA while using the kit method. Results:varying degrees of improvement puerarin three doses of SOD activity and MDA content in mice with viral myocarditis serum,while the expression of CVB3 miRNA has a certain extent. SOD activity and MDA content in serum and impact models process CVB3 miRNA expression in a dosedependent,the high-dose group is better than the low dose group. Conclusion:Puerarin on CVB3-induced mouse model of viral myocarditis CVB3 miRNA expression better inhibition.
