摘要
采用响应面法旨在优化绿色木霉产胶霉菌素培养基。试验选用Min Run Res IV设计对基础培养基的6个影响因子的效应进行评价,筛选显著影响因子,利用最陡爬坡试验确定其取值中心、中心组合试验确定其最佳取值,从而获得优化培养基。Min Run Res IV试验筛选出4个显著性影响因子:葡萄糖、豆粕、磷酸二氢钾和硫酸锌,经过最陡爬坡和中心组合试验,响应面分析得到最佳培养基组合为:葡萄糖26.54 g/L、酵母粉10 g/L、豆粕15.72 g/L、磷酸二氢钾0.452 g/L和硫酸锌0.0582 g/L、硫酸亚铁0.1 g/L,在最佳培养基下胶霉菌素效价较优化前提高了59.71%。
Adopting the response surface method to optimize the medium for gliotoxin produced by Trichoderma viride.We evaluated the effects of 6 factors relating to the basic medium by using Min Run Res IV design,and selected significant impact factors,and determined the midpoint and the optimal value with the steepest ascent design(SAD)and central composite design(CCD),respectively,so as to obtain the optimized medium.Glucose,soymeal,KH2 PO4 and ZnSO4 were the most significant impact factors screened by Min Run Res IV;the response surface analysis by SAD and CCD indicated that:the optimal medium was composed of26.54 g/L glucose,10 g/L yeast powder,15.72 g/L soymeal,0.452 g/L KH2 PO4,0.0582 g/L ZnSO4 and 0.1 g/L FeSO4,which resulted in a 59.71%increase of the titer of gliotoxin compared with the original medium.
