温肾丸对肾阳虚生精障碍大鼠治疗作用的药效学研究 被引量:5

摘要 目的探讨温肾丸对肾阳虚生精障碍大鼠治疗作用的药效学机制。方法选用SPF级SD成年雄性大鼠,体质量180~210g,共60只,随机分为6组:实验组(温肾丸低、中、高剂量组各10只);阳性对照组、模型对照组、空白对照组各10只。除空白对照组外其余各组按腺嘌呤200mg/(kg·d)体质量灌胃10d制备肾阳虚模型。造模结束后开始给药:实验组低、中、高剂量组动物分别给予温肾丸混悬液50mg/kg·d、100mg/kg·d、200mg/kg·d体质量灌胃,连续30d;阳性对照组用克罗米芬混悬液13.5mg/(kg·d)体质量灌胃,连续30d。治疗30d后观察其肾阳虚证表现、血清性激素(T、FSH、LH)、血清抑制素B(INHB)水平变化,并分睾丸组织病理活检及Johnson评分。结果治疗结束后,各药物干预组大鼠肾阳虚症状均有改善;睾丸组织病理显示,模型组可见明显损伤,温肾丸各组及克罗米芬组均有不同程度改善,治疗组及克罗米芬组Johnson评分与模型组比较差异有统计学意义(P<0.01),且温肾丸中、高剂量组优于克罗米芬组,差异有统计学意义(P<0.01);与模型组相比较,温肾丸各剂量组、克罗米芬组大鼠血清中LH和FSH含量降低,T和INHB含量升高,差异有统计学意义(P<0.01),其中温肾丸中、高剂量组与克罗米芬组比较差异有统计学意义(P<0.01)。结论温肾丸能较好地改善模型大鼠肾阳虚症状,调节相关性激素及INHB水平,对腺嘌呤所致大鼠睾丸生精功能障碍模型有明显的治疗作用。 Objective To explore the pharmacodynamic mechanism of Wen Shen Wan in treating kidney-yang deficiency and spermatogenesis disorder in rats. Methods 60 adult male SD rats of SPF grade weighing 180-210g were randomly divided into six groups which were experimental group (10 rats in each low, medium and high dose group of Wen Shen Wan), positive control group, model control group and blank control group (10 rats in each group). Except the blank control group, the other groups were fed with adenine 200 mg/(Kg d) body weight for 10 days to prepare kidney-yang deficiency model. After the model was established, the animals in the low, middle and high dose groups were given Wen Shen Wan suspension 50 mg/kg.d, 100 mg/kg.d and 200 mg/kg.d by intragastric administration for 30 days, while the positive control group was given clomiphene suspension 13.5 mg/kg.d by intragastric administration for 30 days. After 30 days of treatment, the manifestations of kidney-yang deficiency syndrome, changes of serum sex hormones (T, FSH, LH), serum inhibin B (INHB), testicular histopathological biopsy and Johnson score were observed, and the pharmacodynamic mechanism was clarified. Results After treatment, the symptoms of kidney-yang deficiency were improved in each drug intervention group. Testicular histopathology showed that the model group had obvious injury, and the Wen Shen Wan group and clomiphene group there were different degrees of improvement. The Johnson score of the treatment group and clomiphene group there were statistical significantly differences compared with the model group ( P <0.01), and the Wen Shen Wan medium and high dose group was better than clomiphene group. Compared with the model group, the contents of LH and FSH in serum of rats in Wen Shen Wan group and clomiphene group decreased, while the contents of T and INHB increased ( P < 0.01), and the differences between Wen Shen Wan group and clomiphene group were significant ( P <0.01). Conclusion Wen Shen Wan can improve the symptoms of kidney-yang deficiency in rats, regulate the levels of related hormones and INHB, and has a significant therapeutic effect on adenine-induced testicular spermatogenesis dysfunction in rats.
