三黄茵赤汤对大部分肝切除肝衰竭大鼠氧化应激与肝细胞凋亡及再生影响分析

Effects of Sanhuang Yinchi Decoction on Oxidative Stress and Hepatocyte Apoptosis and Regeneration in Most Hepatectomy Rats with Liver Failure

目的:分析三黄茵赤汤对大部分肝切除肝衰竭大鼠氧化应激与肝细胞凋亡及再生影响。方法:选取220只SPF级Wistar大鼠,随机分为正常组、模型组、三黄茵赤汤高、中、低剂量组和促肝细胞生长素组,正常组20只,其它组各40只。建立大部分肝切除肝衰竭大鼠模型,观察各组96小时存活率,检测各组肝功能及肝细胞有丝分裂指数(MI)、肝细胞增殖细胞核抗原(PCNA)指数、超氧化物歧化酶(SOD)、丙二醛(MDA)含量、半胱天冬蛋白酶(caspase-3)表达。结果:三黄茵赤汤各组、促肝细胞生长素组存活率显著高于模型组,差异有统计学意义(P<0.05);三黄茵赤汤各组、促肝细胞生长素组均能显著降低丙氨酸氨基转移酶(ALT)、天冬氨酸转氨酶(AST)及总胆红素(TBIL)含量,增加肝细胞MI及PCNA阳性细胞数,提高SOD活性、降低MDA含量,抑制caspase-3表达,差异有统计学意义(P<0.05);三黄茵赤汤中、低剂量组各指标改善低于促肝细胞生长素组,差异有统计学意义(P<0.05);三黄茵赤汤高剂量组、促肝细胞生长素组效果相当,差异无统计学意义(P>0.05)。结论:三黄茵赤汤能有效减少大部分肝切除肝衰竭大鼠肝细胞凋亡,促进肝细胞再生,提高存活率,其机制可能与三茵赤汤可通过抗氧化应激抑制caspase-3蛋白表达相关。

Objective: The effects of Sanhuang Yinchi Decoction on oxidative stress and hepatocyte apoptosis and regeneration in most hepatectomy rats with liver failure were analyzed. Methods: 220SPF grade Wistar rats were randomly divided into normal group,model group,red soup high,medium and low dose group and hepatocyte growth stimulating hormone group,20in normal group and 40in other groups. The rat model of liver failure was established and the survival rate of 96h was observed. The liver function and mitosis index ( MI),hepatocyte proliferating cell nuclear antigen ( PCNA) index,superoxide dismutase ( SOD),malondialdehyde ( MDA),and cysteine protease ( caspase-3) expression were detected in each group. Results: The survival rate of the Sanhuang Yinchi Decoction and the Hepatocyte Growth Stimulating Hormone group was significantly higher than that in the model group ( P<0. 05). The contents of alanine aminotransferase ( ALT),aspartate aminotransferase ( AST) and total bilirubin ( TBIL) were significantly reduced by the three groups of the Sanhuang Yinchi Decoction group and the Hepatocyte Growth Promoting Factor group,and the liver fine was increased. The number of MI and PCNA positive cells increased the activity of SOD,reduced the content of MDA,inhibited the expression of Caspase-3 ( P<0. 05). The improvement of the indexes of the middle and low dose groups of the Sanhuang Yinchi Decoction group was lower than that of the Hepatocyte Growth Factor group ( P<0. 05),the high dose group and the Growth Promoting Cell Growth of the Sanhuang Yinchi Decoction group. There was no significant difference between the two groups ( P>0. 05). Conclusion: Sanhuang Yinchi Decoction can effectively reduce the hepatocyte apoptosis,promote hepatocyte regeneration and increase the survival rate of liver failure rats. The mechanism may be related to the inhibition of caspase-3protein expression by antioxidation stress.
