摘要
目的:建立一种可现场快速检测空肠弯曲杆菌的方法。方法:应用重组酶聚合酶等温扩增(recombinase polymerase amplification,RPA)技术,设计引物探针,建立侧向流RPA法,使用特异性、灵敏性实验对该方法进行验证。结果:实验所用的所有非空肠弯曲菌在侧向流试剂盒上都是阴性的,空肠弯曲菌都是阳性的;该方法的检出限为1.2×10^3 CFU/mL。结论:本研究制作的RPA技术制作的侧向流试剂盒特异性和灵敏性良好,简单快捷,具有较强的应用价值。
Objective:To establish a rapid method for the detection of Campylobacter jejuni in situ.Methods:Using recombinase polymerase amplification(RPA)technique,primer probes were designed,and Lateral Flow RPA Method was established.Specificity and sensitivity tests were used to verify the method.Result:All non-campylobacter jejuni used in the experiment were negative on the side-flow kit,and Campylobacter jejuni was positive.The detection limit of this method was 1.2×10^3 CFU/mL.Conclusion:The lateral flow kit made by RPA technology in this study has good specificity and sensitivity,simplicity and rapidity,and owns strong application value.
