摘要
目的::研究1-磷酸鞘氨醇(1-phosphate sphingosine,S1P)对牙髓细胞生长、增殖及迁移作用的影响。方法:采用MTT细胞增殖和细胞毒性试验试剂盒检测牙髓细胞(dental pulp cells,DPCs)毒性和细胞增殖,采用划痕实验和Transwell过滤小室过实验评价S1P对牙髓细胞迁移的影响,采用实时荧光定量PCR(qRT-PCR)检测MMP-1、MMP-2和MMP-13 mRNA表达水平。结果:MTT实验结果显示S1P在0.01~1μmol/L浓度范围内对牙髓细胞无毒性作用,划痕实验及Transwell过滤实验结果显示S1P能以剂量依赖的方式影响DPCs的迁移,qRT-PCR实验结果显示S1P显著上调了MMP-2和MMP-13的mRNA。结论:S1P在一定浓度范围内是DPCs迁移的重要启动子,其上调MMP-2和MMP-13也参与其中。S1P可能介导牙髓组织损伤后的创面愈合,可作为牙髓再生治疗的一部分。
Objective:To study the effect of 1-phosphate sphingosine(S1 P)on the growth,proliferation and migration of dental pulp cells.Methods:Cytotoxicity and cell proliferation were detected by MTT cell proliferation and cytotoxicity assay kit.The effect of S1 P on migration of dental pulp cells was evaluated by scratch assay and Transwell filtration chamber assay,and the mRNA levels of MMP-1,MMP-2 and MMP-13 were detected by real-time quantitative polymerase chain reaction(qRT-PCR).Results:S1 P had no toxic effect on dental pulp cells in the concentration range of 0.01~1 μmol/L,and it could affect the migration of DPCs in a dose-dependent manner.Finally,S1 P significantly up-regulated the mRNA expression of MMP-2 and MMP-13.Conclusion:S1 P is an important promoter of DPCs migration within a certain concentration range,and its up-regulation of MMP-2 and MMP-13 is also involved.S1 P may mediate wound healing after pulp tissue injury and can be used as part of pulp regeneration therapy.
