Effect of short-term high-fat diet on intestinal microorganisms in male and female mice

摘要 目的探讨短期高脂饮食对不同性别小鼠肠道微生物的影响。方法将16只C57BL/6J小鼠(雌雄各半)按雌雄各随机分为2组,分别喂食基础饲料(对照组)和45%高脂饲料(高脂组),饮食干预2周后,收集小鼠的新鲜粪便样本,提取粪便细菌总基因组DNA,对其16S rDNA的V3+V4区域进行扩增及测序,分析菌群的组成及丰富度变化。结果高脂饮食改变了雌性和雄性小鼠肠道微生物组成及物种多样性。与相应对照组比较,雌性高脂组小鼠厚壁菌门及该菌门中Leptum菌种的相对丰富度均显著下降(P﹤0. 05),雄性高脂组差异均无统计学意义(P>0.05)。结论短期高脂饮食对雌性和雄性小鼠肠道微生物组成及结构有不同的影响,可能与不同性激素与肠道菌群的相互作用有关。 Objective To investigate the effect of high-fat diet on intestinal microorganisms in male and female mice. Methods Eight male and eight female C57BL / 6J mice were divided into two groups separately. The mice in control groups were fed with normal diet,while those in test group with a 45 % high-fat diet. After dietary intervention for 2 weeks,fresh fecal samples were collected from each group,from which fecal bacterial genomic DNA was extracted for PCR amplification based on the V3 + V4 region of 16S-rDNA and sequencing. The changes in composition and relative abundance of gut microbiota were analyzed. Results High-fat diet changed the composition and diversity of intestinal microorganisms in male and female mice. Compared with those in the corresponding control groups,the relative abundances of Firmicutes and Leptum specie in female mice in test groups decreased significantly (P﹤0. 05),while those in male mice showed no significant difference (P > 0. 05). Conclusion Short term high-fat diet showed different effects on the composition and structure of intestinal microorganisms in female and male mice,which might be associated with the interaction of sex hormones with intestinal microorganisms.
