摘要
目的探讨锦红汤对脓毒症小鼠血小板聚集和分泌功能的影响。方法选择健康清洁级C57BL/6雄性小鼠分为假手术组、盲肠结扎组和锦红汤组,检测各组术后4 h和72 h的血小板计数、血小板聚集率、P-选择素表达率和五羟色胺(5-HT)血清浓度。结果术后4 h,盲肠结扎组和锦红汤组的血小板计数较假手术组均明显降低(×10^9/L:508.00±98.42、481.20±38.41vs.784.00±85.97,P<0.05);术后72 h,锦红汤组较盲肠结扎组明显升高(×10^9/L:574.80±78.71 vs.461.00±64.77,P<0.05)。术后4 h,盲肠结扎组血小板聚集率较假手术组明显升高[(56.67±0.58)%vs.(41.33±1.53)%,P<0.05],锦红汤组[(48.00±3.46)%]较盲肠结扎组明显降低(P<0.05)。术后72 h,盲肠结扎组血小板聚集率较假手术组明显降低[(12.67±4.04)%vs.(43.33±2.89)%,P<0.05),锦红汤组[(29.00±5.29)%]较盲肠结扎组明显升高(P<0.05),但仍低于假手术组(P<0.05)。术后4 h,盲肠结扎组P-选择素表达率较假手术组明显升高[(15.53±1.60)%vs.(4.01±1.05)%,P<0.05],锦红汤组[(13.63±1.18)%]较盲肠结扎组明显降低(P<0.05),但仍高于假手术组(P<0.05)。术后72 h,盲肠结扎组P-选择素表达率较假手术组明显升高[(16.78±1.60)%vs.(2.33±0.14)%,P<0.05],锦红汤组[(12.92±2.47)%]较盲肠结扎组明显降低(P<0.05),但仍高于假手术组(P<0.05)。盲肠结扎组和锦红汤组术后4 h和72 h 5-HT水平均低于假手术组(P<0.05),但盲肠结扎组和锦红汤组比较差异无统计学意义(P>0.05)。结论锦红汤能改善脓毒症小鼠血小板数量和聚集能力,通过下调P-选择素和α颗粒分泌发挥抗炎作用。
Objective To investigate the effect of Jinhong Decoction on platelet aggregation and secretion in mice with sepsis. Methods C57BL/6 male mice were divided into sham operation group, cecal ligation group and Jinhong Decoction group. Platelet count, platelet aggregation rate, serotonin (5-HT) serum concentration and P 一 selectin expression rate were detected at 4 h and 72 h after operation. Results Four hours after operation, the platelet counts of the cecal ligation group (508. 00 ± 98. 42) x 10^9/L and Jinhong Decoction group(481.20 ± 38. 41 ) x 10^9/L were significantly lower than those of the sham operation group ( 784. 00 ± 85. 97 ) x 10^9/L ( P<0. 05). Seventy-two hours after operation, the platelet count of Jinhong Decoction group( 574. 80 ± 78. 71 ) x 10^9/L was significantly higher than that of the cecal ligation group (461.00 ± 64. 77 ) x 10^9/L ( P< 0. 05 ). Four hours after operation, the platelet aggregation rate in the cecal ligation group was ( 56. 67 ± 0. 58 )%, which was significantly higher than that in the sham operation group (41.33 ± 1.53 )%( P < 0. 05 ). The platelet aggregation rate of Jinhong Decoction group (48. 00 ± 3. 46 )% was significantly lower than that of the cecal ligation group ( P< 0. 05 ). At 72 h postoperatively, the platelet aggregation rate in the cecal ligation group was ( 12. 67 ± 4. 04 )%, which was significantly lower than that in the sham operation group (43. 33 ±2. 89 )%( P<0. 05 ). The platelet aggregation rate in Jinhong Decoction group (29. 00 ± 5. 29)% was significantly higher than that in the cecal ligation group ( P < 0. 05 ), but still lower than that in the sham operation group ( P < 0. 05). Four hours after operation, the expression rate of P-selectin in the cecal ligation group was ( 15. 53 ± 1.60 )%, which was significantly higher than that in the sham operation group (4.01%± 1.05 )%( P <0. 05). The expression rate of P - selectin in Jinhong Decoction group (13.63 ±1.18)% was significantly lower than that in the cecal ligation group (P <0. 05), but still higher than the sham operation group ( P < 0. 05 ). At 72 h postoperatively, the expression rate of P - selectin in the cecal ligation group was (16. 78 ± 1.60)%, which was significantly higher than that in the sham operation group (2. 33 ±0. 14)%(P <0. 05). The expression rate of P - selectin in Jinhong Decoction group ( 12. 92 ± 2. 47 )% was significantly lower than the cecal ligation group ( P <0. 05), but still higher than that in the sham operation group ( P <0. 05 ). The 5 一 HT levels of the cecal ligation group and the Jinhong Decoltion group were both lower than those of the sham operation group at 4 hours and 72 hours after opperation, with statistically significant differences ( P < 0. 05), but there was no statisitically significant difference between the cecal ligation group and the Jinhorg Decoction group( P > 0. 05 ). Conclusion Jinhong Decoction can improve the platelet number and aggregation ability in sepsis, and exert anti-inflammatory effect by down-regulating the secretion of P - selectin and α-particle.
