摘要
目的观察评估改进的复方地芬诺酯大鼠便秘模型的造模效果、可重复性和安全性。方法雄性SD大鼠18只,4~6周龄,适应性饲养1周后,按照体质量区组随机分成对照组、模型组和治疗组3组。模型组和治疗组按体质量以10 mL/kg灌胃1.5 mg/mL复方地芬诺酯混悬液,1次/d,连续灌胃14 d;造模第8天开始治疗组大鼠在造模灌胃基础上按体质量以10 mL/kg灌胃0.05 mg/mL乐可舒混悬液,1次/d,连续灌胃7 d。观察大鼠造模前、造模7 d和14 d以下指标:一般情况,整体状况、体质量、摄食量、饮水量和尿量;24 h的粪便粒数;24 h的粪便重量;粪便含水率;首次排便时间。重复进行上述实验3次。结果 3批次大鼠造模7 d和造模14 d后粪便粒数减少、粪便重量减轻、含水率下降,分别与造模前比较,差异均有统计学意义(P<0.05),造模7 d与造模14 d的差异均无统计学意义(P>0.05);造模7 d和造模14 d,大鼠的首次排便时间均比同批次对照组延长,差异均有统计学意义;治疗组干预后粪便粒数增多、粪便重量增加、含水率上升,与造模7 d的差异均有统计学意义。结论改进的复方地芬诺酯大鼠便秘模型是一种相对简便的功能性便秘大鼠模型,具有较好的可重复性和安全性。
Objective Observation and evaluation of model effect, repeatability and safety of the compound Difenolone in the model of constipation. Methods 18 male SD rats, 4-5 weeks old,were divided into control group,model group and treatment group with randomized block according to weight after one week of adaptive feeding.The model group and treatment group were given a gavage of 1.5 mg/mL compound difenolate by weight 10 mL/kg one time every day and for 14 days. Beginning with the 8 th day, treatment group were given a gavage of 0.5 mg/mL Dulcolax by weight 10 mL/kg one time every day and for 7 days. The indexes were observed the 0 th, 7 th and 14 th day including: Overall status, weight, food intake, water intake and urine output, fecal number for 24 h, fecal weight for 24 h, water content of feces and first time defecation. The above experiment was repeated 3 times. Results In the same batch of rats in model group, the fecal number decreased, fecal weight decreased and water content decreased on the 7 th and 14 th day. Comparison with pre-molding, the differences within the group were statistically significant(P < 0.05).The differences of constipation model rats between 7 th and 14 th day were not statistically significant(P > 0.05). There were longer first time defecation in constipation model rats than control group on the 7 th and 14 th day. The differences between the two groups were statistically significant. In the treatment group, the fecal number decreased, fecal weight decreased and water content decreased on the 14 th day. The differences between the 7 th and14 th day were statistically significant. Conclusions The improved constipation model is a relatively simple model of functional constipationmodel of rats with compound difenolate, which is repeatable and safe.
