红景天苷抑制哮喘小鼠气道重塑及其机制研究 被引量:13

Inhibitory effect of salidroside on airway remodeling and its mechanism in asthmatic mice

摘要 目的研究红景天苷抑制哮喘小鼠气道重塑及对核因子-κB/转化生长因子-β1(NF-κB/TGF-β1)信号通路的影响。方法将60只C57bL/6小鼠随机分为对照组、模型组和高、低剂量实验组,每组15只。模型组和实验组通过腹腔注射、雾化吸入卵清蛋白构建哮喘小鼠,对照组给予等量生理盐水处理。高、低剂量实验组分别于卵清蛋白雾化吸入的同时灌胃给予50,100 mg·kg^-1红景天苷,模型组和对照组给予等量生理盐水。检测小鼠支气管肺泡灌洗液(BALF)中炎症细胞水平;以苏木精-伊红(HE)、过碘酸雪夫(PAS)和Masson染色法观察小鼠肺组织病理形态,以逆转录聚合酶链式反应(RT-PCR)法检测小鼠肺组织中NF-κB/TGF-β1通路mRNA表达情况。结果对照组、模型组和低、高剂量实验组肺组织中NF-κB P65 mRNA相对表达量分别为0. 24±0. 06,0. 96±0. 21,0. 45±0. 07,0. 42±0. 06,TGF-β1 mRNA相对表达量分别0. 35±0. 04,1. 05±0. 21,0. 63±0. 05,0. 62±0. 05,与模型组比较,差异均有统计学意义(均P <0. 05)。与对照组比较,模型组小鼠BALF中淋巴细胞、巨噬细胞、中性粒细胞及嗜酸性粒细胞数量均明显升高(均P <0. 05),但实验组小鼠BALF中炎症细胞数量均低于模型组(P <0. 05)。结论红景天苷早期干预可以通过抑制NF-κB/TGF-β1信号通路活化,降低哮喘小鼠慢性呼吸道炎症程度及气道重塑。 Objective To investigate the inhibitory effect of salidroside on airway remodeling and its influence on nuclear factor -κB /transforming growth factor -β1( NF -κB /TGF -β1) signaling pathway in asthmatic mice. Methods Sixty C57bL /6 mice were randomly divided into control group,model group and low,high dose experiment groups,15 mice in each group. The asthmatic mice in model group and experiment groups were constructed by intraperitoneal injection and atomization inhalation of ovalbumin,and control group was given the same amount of normal saline. Low,high dose experiment groups were intragastric given 50, 100 mg·kg^-1 salidroside at the same time of ovalbumin atomization inhalation,model group and control group were given the same amount of normal saline. The levels of inflammatory cells in bronchoalveolar lavage fluid ( BALF) of mice were detected;hematoxylin - eosin ( HE),Periodic acid Schiff ( PAS) and Masson staining were used to observe the pathological morphology of lung tissue in mice,and the expression of NF -κB /TGF -β1 pathway mRNA in lung tissue of mice was detected by reserve transcription - polymerase chain reaction ( RT - PCR). Results The relative expression levels of NF -κB P65 mRNA in lung tissue of control group,model group and low,high dose experiment groups were 0. 24 ± 0. 06,0. 96 ± 0. 21,0. 45 ± 0. 07,0. 42 ± 0. 06 and the relative expression levels of TGF -β1 mRNA were 0. 35 ± 0. 04,1. 05 ± 0. 21,0. 63 ± 0. 05,0. 62 ± 0. 05,there were significant difference compared with model group( all P < 0. 05). Compared with control group,the number of lymphocytes,macrophages,neutrophils and eosinophils in BALF in model group increased significantly ( P < 0. 05),but the number of inflammatory cells in BALF of experiment group were lower than those of model group ( P < 0. 05). Conclusion Salidroside early intervention can reduce the degree of chronic respiratory inflammation and airway remodeling in asthmatic mice by inhibiting the activation of NF-κB /TGF -β1 signaling pathway.
