糖尿病心肌纤维化对大鼠心肌间质telocyte细胞影响的研究

摘要 目的探究糖尿病大鼠心肌组织纤维化对心肌间质中telocyte细胞数量变化的影响。方法40只大鼠随机分为对照组(Con)和T2DM组,每组各20只。T2DM组腹腔注射STZ(27.5mg/kg)建立大鼠糖尿病模型,建模成功后以高脂高糖饮食饲养20周。胶原蛋白免疫组织化学、天狼猩红、Masson三色染色鉴定心肌纤维化程度。免疫荧光技术标记出心肌间质telocyte细胞后,计数心肌间质telocyte细胞与心肌细胞数量相对比值,比较两组心肌间质telocyte细胞的数量差异。结果与Con组比较,T2DM组FBG、摄食量、饮水量、尿量均升高,天狼猩红染色结果显示,心肌组织纤维化程度加深[(4989.00±340.80)vs(8241.00±1930.90)],心肌组织Ⅰ型胶原蛋白、Ⅲ型胶原蛋白升高[(892±38)vs(5381±634),(2000±256)vs(11658±658)];心肌间质telocyte细胞数量在心肌组织细胞总量中所占的比例减少[(0.052±0.010)vs(0.024±0.003)](P<0.05)。结论伴随糖尿病大鼠心肌纤维的发生,心肌间质telocyte细胞数量减少。 Objective To investigate the effect of diabetic myocardial fibrosis on myocardial interstitial telocyte cells in diabetic rats.Methods A total of 40 rats were selected in this study and divided into two groups:Con group and type 2 diabetes mellitus(T2DM)group.T2DM rat model was establishment by intraperitoneal injection of streptozotocin(STZ)27.5 mg/kg.Then the T2DM group was treated with high-fat and high-sugar diet for 20 weeks.The degree of the myocardium fibrosis was evaluated by immunohistochemistry of collagen,sirius red staining,and Masson staining.Myocardial interstitial telocyte cells were labeled with immunofluorescence techniques.The relative number of myocardial interstitial telocyte cells was calculated,and compared between T2DM group and Con group.Results FBG,food and water intake,and urine volume were all increased in T2DM group than in Con group.Sirius red staining showed that the degree of the myocardium fibrosis was more severe[(8241.00±1930.90)vs(4989.00±340.80)],collagenⅠ[(5381±634)vs(892±38)]and collagenⅢ[(11658±658)vs(2000±256)]were higher,while the percentage of myocardial interstitial telocyte cells in myocardial cells were decreased[(0.024±0.003)vs(0.052±0.010)]in T2DM group than in Con group.Conclusion With the progress of myocardial fibrosis in diabetic rats,the number of myocardial interstitial telocyte cells decreased.
