夏枯草提取物对自发性高血压大鼠血压及血管活性物质的影响 被引量:25

Effects of Prunella Vulgaris L. Extract onBloodPressure and Vascular ActiveSubstance in Spontaneously Hypertensive Rats

摘要 目的:观察夏枯草提取物对自发性高血压大鼠血压的控制情况及相关指标的变化。方法:对照组为10只WKY大鼠,并将筛选出的40只自发性高血压大鼠(SHR)随机分为:模型组、阳性药组、夏枯草提取物高剂量组、夏枯草提取物低剂量组,每组10只。从14周龄时起给药,1次/d,连续灌胃5周。每周测量大鼠体质量。同时收集大鼠收缩压和行为学指标,给药5周后取材。运用ELISA检测大鼠给药5周后血清ANGⅡ、NE、ET、CGRP、TXB2的水平。结果:与模型组相比,阳性药组、夏枯草提取物高剂量组、夏枯草提取物低剂量组对收缩压均有一定程度的降低(P<0.05),其中,夏枯草提取物高剂量组的降压效果优于夏枯草提取物低剂量组,阳性药组无显著变化(P>0.05);随给药时间的延长,旋转耐受时间、易激惹程度评分均有不同程度的改善(P<0.05),阳性药组无显著变化(P>0.05),进行夏枯草提取物干预5周后,与模型组相比,阳性药组、夏枯草提取物高剂量组血清ANGⅡ浓度低于模型组(P<0.05);各组血清NE、ET、TXB2浓度均低于模型组(P<0.05),CGRP浓度均高于模型组(P<0.05)。结论:夏枯草提取物降压的作用机理可能与降低血清中、NE、ET含量,增加血清中CGRP含量相关,且存在量效关系。 Objective:To observe the control of Prunella vulgaris L. extract on blood pressure and the changes of related indexes in spontaneously hypertensive rats. Methods: Ten WKY rats in the control group were randomly divided into 40 spontaneously hypertensive rats(SHR): model group, positive drug group, Prunella vulgaris extract high dose group, Prunella vulgaris extract low dose group, 10 in each group. The drug was administered from the age of 14 weeks, once a day, and continuously administered for 5 weeks. Rat body weight was measured weekly. At the same time, the systolic blood pressure and behavioral indexes of the rats were collected and taken out after 5 weeks administration. The levels of serum ANGII, NE, ET, CGRP and TXB2 were measured by ELISA for 5 weeks after administration. Results: Compared with the model group, the positive drug group, the high-dose and low-dose groups of Prunella vulgaris L. had a certain degree of reduction in systolic blood pressure(P<0.05), among which the high dose of Prunella vulgaris L. extract had better antihypertensive effect thanthe low dose group of Prunella vulgaris L. extract, and there was no significant change in the positive drug group(P>0.05). With the prolongation of the administration time, the rotation tolerance time and irritability scores were improved to different extents(P<0.05). There was no significant change in the positive drug group(P>0.05). After 5 weeks intervention with Prunella vulgaris L. extract, the serum ANGII concentration of the positive drug group and the high dose group of Prunella vulgaris L. extract was lower than that of the model group. The concentrations of NE, ET and TXB2 in each group were lower than that in the model group(P<0.05), and the concentration of CGRP was higher than that in the model group(P<0.05). Conclusion: The mechanism of antihypertensive effect of Prunella vulgaris L. extract may be related to reducing serum NE and ET contents and increasing serum CGRP content, and there is a dose-effect relationship.
