摘要
目的:为探讨大黄素对bmSNI模型大鼠腰膨大p-JNK3蛋白是否有影响。方法:50只150~220 g健康雄性SD大鼠随机平均分为正常组(C)、假手术组(SH)、模型组(M)、大黄素组(D)和SP600125组(SP)共5组,通过剪断大鼠双侧胫神经造成bmSNI模型,后2组在手术完毕后即分别予大黄素40 mg/(kg·d)灌胃14 d、SP600125(20μL/d,0.25 g/L)鞘内注射7 d,且于造模前1、后1、2、3、5、7、10、14 d测机械痛敏和热痛敏,每组在造模后7 d取4只取材腰膨大,对腰膨大进行p-JNK3蛋白免疫印迹检验比较各组差异。结果:M组术后第7天机械敏最敏感。D组术后第2天机械痛阈值显著低于M组,到术后3 d达最低值,此后逐渐上升,机械痛阈值均显著高于M组。结论:大黄素对于NP有明显疗效,且强于JNK抑制剂SP600125。然而术后各组腰膨大Western Blotting检测无显著差异,表明大黄素可能通过其他途径治疗神经病理性疼痛(NP)。上述结论尚待进一步开展更多大样本、高质量研究予证实。
Objective:To investigate whether Emodin has an effect on p-JNK3 protein in lumbar enlargement of bmSNI model rats. Methods:Fifty 150~220 g healthy male Sprague-Dawley rats were randomly divided into normal group(C), sham operation group(SH), model group(M), Emodin group(D) and SP600125 group(SP). The bmSNI model was induced by cutting the bilateral tibial nerves in rats. D group were given Emodin 40 mg/(kg·d) by gavagefor 14 days. SP group were given SP600125(20 L/d, 0.25 g/L) intrathecal administration for 7 days. One day before the operation, and on 1,2,3,5,7,10,14 d after operation,we measured the rats’ mechanical and thermal hyperalgesia. On 7 d after modeling,4 rats of each group were taken out for obtaining the lumbar enlargement samples, and these tissues were taken tested by JNK3 blotting. Results:M group were mostly sensitive to mechanical sensitivity on 7 d after surgery, while D group showed significantly lower sensitivity than that of the M group on the 2 nd day after surgery, and reached the lowest value 3 days after operation. From the 4 th to 14 th day, mechanical sensitivity increased gradually in D group, which was significantly higher than that of the M group. Conclusion:Emodin is effective for neuropathic pain of bmSNI model and stronger than JNK inhibitor SP600125. However, Western blotting analysis of lumbar enlargement in each group showed no significant difference among the groups. This study shows that Emodin may have effect on the bmSNI model rats through otherways. The above conclusions need to be further confirmed by more large-scale, high-quality research.
