白藜芦醇对乳腺癌易感基因FBXO10启动子及FBXO10 mRNA表达的影响

摘要 目的探讨白藜芦醇对乳腺癌易感基因FBXO10启动子及FBXO10mRNA表达的影响。方法采用瞬时转染法将不同长度、不同乳腺癌易感性(WF)及抗性(Wky)FBXO10启动子报告质粒转染入T细胞来源细胞系Jurkat细胞,然后加入白藜芦醇孵育,用荧光素酶检测仪检测各组相对荧光素酶活性。将长度4.2kbWF和WkyFBXO10启动子报告质粒转染入Jurkat细胞,分别加入浓度0、10、25、50、100μmol/L的白藜芦醇,通过荧光素酶检测仪检测每组的相对荧光素酶活性。实时定量PCR检测不同浓度白藜芦醇处理的Jurkat细胞FBXO10mRNA表达。结果荧光素酶检测仪的相对荧光素酶活性结果显示白藜芦醇抑制Jurkat细胞中不同长度WF和WkyFBXO10启动子的活性,并且浓度越高,对4.2kbWF和WkyFBXO10启动子的表达的抑制越强(P均<0.05);实时荧光定量PCR结果显示,不同浓度的白藜芦醇可抑制Jurkat细胞中FBXO10mRNA的表达,浓度越高,抑制作用越强(P均<0.05)。结论白藜芦醇能够抑制乳腺癌易感基因FBXO10启动子的活性并且降低FBXO10mRNA表达。 Objective To investigate the effects of resveratrol on the breast cancer susceptibility gene FBXO10 promoter and FBXO10 mRNA expression.Methods The susceptible WF and resistant Wky FBXO10 promoter reporter plasmids of different lengths were transfected into the T cell line Jurkat by transient transfection.Resveratrol was added,and the relative luciferase activity of each group was detected by luciferase assay.The 4.2 kb susceptible WF and resistant Wky FBXO10 promoter reporter plasmids were transfected into Jurkat cells,and resveratrol at different concentrations(0,10,25,50,and 100μmol/L)was added,and the relative luciferase activity of each group was detected by luciferase assay.The real-time quantitative PCR was used to detect the expression of FBXO10 mRNA in Jurkat cells treated with different concentrations of resveratrol.Results Resveratrol inhibited the expression of WF and Wky FBXO10 promoters of different lengths in Jurkat cells as shown in luciferase assay,and the higher the concentration,the stronger the inhibitory effect on the expression of the 4.2 kb WF and Wky FBXO10 promoters(all P<0.05);real-time quantitative PCR showed that different concentrations of resveratrol could inhibit the expression of FBXO10 mRNA in Jurkat cells,and the higher the concentration,the stronger the inhibition(all P<0.05).Conclusion Resveratrol can inhibit the activity of breast cancer susceptibility gene FBXO10 promoter and decrease FBXO10 mRNA expression.
