桃红四物汤对UV辐射致人真皮微血管内皮损伤后TGF-β、Smad2/3表达影响的实验研究 被引量:5

摘要 目的:旨在研究桃红四物汤对模拟UVA照射后损伤HDMEC的保护作用。方法:以5J/cm2UVA辐射HDMEC建立紫外线光损伤的人真皮微血管内皮细胞模型,分别给予不同浓度的桃红四物汤,检测不同浓度药物作用下的细胞凋亡及分泌TGF-β、Smad2/3蛋白的情况。结果:与空白对照组比较,模型对照组细胞凋亡数显著上升,细胞培养上清液中TGF-β含量降低,细胞TGF-β、Smad2/3表达降低;与模型对照组比较,桃红四物汤含药血清干预的各组细胞凋亡数均显著下调,培养上清液中TGF-β含量增加,细胞TGF-β、Smad2/3表达升高,以高剂量组和阳性对照组上升最为明显。结论:桃红四物汤含药血清能有效保护UVA诱导的真皮微血管内皮细胞损伤,其机制可能与激活TGF-β/Smad信号通路有关。 Objective:The aim of this study was to investigate the protective effect of Taohong Siwu decoction on damaged HDMEC induced after simulated UVA irradiation.Methods:5J/cm^2 UVA radiation HDMEC was used to establish a Human Dermal Microvascular Endothelial Cell model damaged by ultraviolet light.The effects of different concentrations of Taohong Siwu decoction on cell apoptosis and secretion of TGF-βand Smad2/3 proteins were detected.Results:Compared with the blank control group,the number of cell apoptosis in the model control group increased significantly,the content of TGF-βin the supernatant of cell culture decreased,and the expression of TGF-βand Smad2/3 decreased in each group.Compared with the model control group,cell apoptosis was significantly down-regulated in all groups treated with Taohong Siwu decoction containing drug serum,TGF-βcontent was increased in cell culture supernatant,and the expression of TGF-βand smad2/3 was increased in all groups,especially in the high-dose group and the positive control group.Conclusion:Taohong Siwu decoction containing drug serum can effectively protect UVA-induced damage of dermal microvascular endothelial cells,and its mechanism may be related to the activation of TGF-β/Smad signaling pathway.
