表柔比星通过P53、Bcl-2信号通路诱导骨髓间充质干细胞凋亡

Epirubicin caused apoptosis of bone mesenchymal stem cells via activating P53 and Bcl-2 signaling pathway

摘要 目的探讨表柔比星是否会引起骨髓间充质干细胞的凋亡,进而加重其骨髓抑制作用。方法骨髓间充质干细胞中加入表柔比星培养48 h,应用MTT实验、透射电镜实验和Western blot实验研究表柔比星是否可以诱导骨髓间充质干细胞凋亡。结果表柔比星降低了BMSCs的活性,诱导了BMSCs凋亡。表柔比星处理后,骨髓间充质干细胞的P-P53、P53蛋白表达增加,Bcl-2蛋白表达下降。结论表柔比星可以通过激活P53、Bcl-2信号通路引起骨髓间充质干细胞的凋亡。 Objective To investigate if epirubicin causes the apoptosis of BMSCs and in turn aggravate its myelosuppression. Methods BMSCs were exposed to epirubicin for 48 h,and cell apoptosis was determined by MTT,transmission electron microscope and Western blot. Results Epirubicin reduced the survival ratio and caused the apoptosis of BMSCs. The phosphorylated P53 and total P53 proteins were increased in epirubicin treated BMSCs. Antiapoptotic Bcl-2 protein was reduced in epirubicin treated BMSCs. Conclusion Apoptosis of bone marrow mesenchymal stem cells caused by epirubicin via P53 and Bcl-2 signaling pathway.
