摘要
目的:研究补阳温肾合剂对肾阳虚大鼠模型下丘脑—垂体—肾上腺皮质轴(HPA轴)的影响,探讨补阳温肾合剂作用机理。方法:将50只雄性Sprague-Dawley大鼠随机分为正常对照组、模型组、低剂量组、中剂量组、高剂量组,每组10只。除正常对照组外,其余大鼠后肢肌注氢化可的松,建立肾阳虚模型。造模成功后,每组大鼠分别予以不同药物连续灌胃28天,第29天处死大鼠,测量并比较各组血清T、ACTH、CORT水平及肾上腺、垂体、下丘脑重量。结果:造模后结果与正常对照组相比,大鼠血清T、ACTH、CORT含量、肾上腺、垂体、下丘脑重量均明显降低(P<0.05);药物干预后,与模型组相比,低、中、高剂量组大鼠血清T水平均有提升(P<0.05),中、高剂量组可显著提高ACTH水平(P<0.05),低、中剂量组对CORT水平提升无明显效果,高剂量组能一定程度提高CORT水平(P<0.05)。相比正常对照组,模型组大鼠体重增加缓慢;与模型组相比,低、中、高剂量组大鼠体质量变化均有不同程度增加(P<0.05),对于肾上腺、垂体、下丘脑重量低剂量组改善不明显,高剂量组疗效优于中剂量组(P<0.05)。结论:补阳温肾合剂对肾阳虚大鼠模型HPA轴具有良好的调节作用,其剂量对肾阳虚体质的改善程度有一定影响。
Objective:to study the effect of Buyang Wenshen Oral Liquid on hypothalamus-pituitary-adrenal cortex axis(HPA axis) in rats with deficiency of kidney yang,and to explore the mechanism of Buyang Wenshen Oral Liquid.Methods:Fifty male SPF grade SD rats were randomLy divided into normal control group,model group,low dose group,middle dose group and high dose group with 10 rats in each group.The model of kidney yang deficiency was established by intramuscular injection of hydrocortisone into the hind limbs of rats except the normal control group.After successful modeling,the rats in each group were perfused with different drugs for 28 days,and the rats were killed on the 29 th day.The serum levels of T、ACTH、CORT and the weights of adrenal gland,pituitary gland and hypothalamus were detected and compared in each group.Results:Compared with the normal control group,the content of serum T、ACTH、CORT,the weight of adrenal gland,pituitary and hypothalamus were significantly decreased(P< 0.05).Compared with the model group,the serum T level in the model group,middle dose group and high dose group increased significantly(P< 0.05).The level of ACTH was significantly increased in middle and high dose groups(P< 0.05).The low and middle dose groups had no obvious effect on the level of CORT,but the high dose group could increase the level of CORT to some extent(P< 0.05).) Compared with the normal control group,the weight gain of the model group was slower than that of the model group.The changes of body mass in low,medium and high dose groups were all increased in varying degrees(P< 0.05).For adrenal gland,pituitary gland and hypothalamus,the effect of high dose group was better than that of middle dose group(P< 0.05),but the effect of high dose group was better than that of middle dose group(P<0.05).Conclusion:Buyang Wenshen oral liquid has a good regulating effect on the HPA axis of kidney yang deficiency rat model,and its dosage has certain influence on the improvement degree of kidney yang deficiency constitution.
