腹水Walker256乳腺癌细胞接种建立SD大鼠胫骨骨癌痛模型 被引量:1

Establishment of SD Rat Model with Tibial Cancer Pain by Ascites Cultured Walker256 Breast Cancer Cells Inoculation

摘要 探讨应用腹水传代培养的Walker256大鼠乳腺癌细胞系建立SD大鼠胫骨癌痛模型,为骨癌痛的研究和镇痛药物开发创造有利条件.结果显示,在骨癌模型组,接种后体质量呈缓慢下降,从第15天起与接种前相比存在极显著差异,到第21天时,体质量下降了12. 5%(P <0. 001);热缩足潜伏期也逐渐下降,从第6天开始,与接种前相比存在显著性差异,至第21天时,缩足潜伏期下降了34. 1%,接种的对侧后足也出现类似现象;X-影像观察显示骨癌痛模型大鼠的胫骨结构受到破坏,骨密度不均一.以上结果表明,应用腹水传代的Walker256乳腺癌细胞接种后能产生较为稳定的痛觉过敏等骨癌痛行为,SD大鼠胫骨癌痛模型建立成功. To investigate the possibility of ascites cultured Walker256 breast cancer cells line to establish the model of tibial cancer pain in SD rats,to create favorable conditions for the study of bone cancer pain and the development of analgesic drugs. The results showed that in the model group of bone cancer,the weight decreased slowly after inoculation,there was a significant difference from the 15 th day to the pre-inoculation,and the body mass was decreased by 12. 5% on the21 th day( P 0. 001). The heat paw withdrawal latency was shortened gradually,too,there was evident difference from the 6 th day to the pre-inoculation,and the paw withdrawal latency was decreased by 34. 1% on the 21 th day( P 0. 001).The contralateral foot of inoculation showed a similar phenomenon. Radiograph data showed that the tibial bone structure of the model rats was obviously destroyed,and bone density was not uniform. The results illustrated that inoculation with ascites cultured Walker256 breast cancer cells could produce stable bone cancer pain behavior,such as hyperalgesia,the model of tibial bone cancer pain in SD rats was established successfully.
