连续离子交换技术高效纯化莽草酸

摘要 目的研究利用连续离子交换技术高效纯化发酵液中的莽草酸。方法采用连续离子交换技术对莽草酸预处理液进行阴离子交换树脂预纯化。充分利用预纯化过程中柱流出液的pH和电导率变化与流出液组分变化之间的高度对应关系,对预纯化过程实行精准自动控制。预纯化液再进行阴离子交换树脂精纯、浓缩、喷雾干燥可得到高纯度莽草酸成品。结果采用预纯化工艺去除了预处理液中大量存在的强结合力杂质阴离子,进而大幅提高了精纯步骤的处理量,树脂总使用量减少了约80%,精纯洗脱液色谱纯度高达99%以上,洗脱收率提高了15%以上。结论该工艺所得产品纯度高,不需要结晶操作,实现了生产过程的连续化、自动化,大幅提高了生产效率,降低了生产成本,适合大规模工业化生产。 Objective The highly efficient purification of shikimic acid in fermentation broth was studied through continuous ion exchange technology. Methods The continuous ion exchange technology was applied to pre-purify the shikimic acid pretreated liquid, which made full advantage of the relationship of the change of pH and conductivity with the change of the component of elute to realize the continuous and automatic production process. Then the obtained pre-purified liquid would be purified further by anion exchange resin refining step, after concentrating and spray-drying, the final shikimic acid product with high quality has been obtained. Results The pre-purifying step has removed plenty of anion impurities with strong bond strength, improved the processing capacity of the refining step greatly. The total demand of the resin has reduced by 80%, the HPLC purity of the eluent of the refining step has reached more than 99%, and the eluting yield of the refining step were increased more than 15%. Conclusion The final product of this purification process has high purity, and no crystallization process was needed. The process improved the productivity significantly and reduced the cost greatly, so it was suitable for commercialscale production.
