子宫内膜异位症内膜组织中HOXA10基因异常DNA甲基化与意义研究

Abnormal DNA methylation of HOXA10 gene in endometriosis and its significance

目的探讨子宫内膜异位症(EMs)内膜组织中HOXA10基因异常DNA甲基化与意义。方法前瞻性选取2016年1月至2018年1月新疆医科大学第一附属医院收治的EMs在位内膜患者28例、EMs异位内膜患者28例,正常对照子宫内膜患者28例。比较三组HOXA10基因启动F1甲基化、F2甲基化和F3甲基化状态、总甲基化率和不孕患者甲基化率。结果EMs在位内膜组与EMs异位内膜组的HOXA10基因启动子甲基化状态无显著差异(P>0.05),EMs在位内膜组、EMs异位内膜组的HOXA10基因启动子甲基化状态高于正常内膜组(P<0.05);EMs在位内膜组与EMs异位内膜组的总甲基化率和不孕患者甲基化率无显著差异,EMs在位内膜组、EMs异位内膜组的总甲基化率和不孕患者甲基化率显著高于正常内膜组(P<0.05)。结论HOXA10基因在EMs中出现HOXA10基因异常DNA甲基化,同时HOXA10基因的DNA甲基化在EMs合并不孕患者内膜组织中的表达能力显著高于非EMs不孕患者。

Objective To investigate the abnormal DNA methylation and significance of HOXA10 gene in endometrial tissue of endometriosis(EMs).Methods 28 patients with EMs eutopic endometrium,28 patients with EMs ectopic endometrium,and 28 normal control endometrial patients were enrolled in the First Affiliated Hospital of Xinjiang University from January to February 2018.Three groups of HOXA10 genes were compared to initiate F1 methylation,F2 methylation and F3 methylation status,total methylation rate,and methylation rate in infertile patients.Results There was no significant difference in the methylation status of the HOXA10 gene promoter between the eutopic endometrium and the EMs ectopic endometrium.The methylation status of the HOXA10 gene promoter in the EMs eutopic endometrium and EMs ectopic endometrium was high.In the normal endometrium group(P<0.05).There was no significant difference between the total methylation rate of EMs in the eutopic endometrium group and the EMs ectopic endometrium group and the methylation rate of infertile patients.EMs eutopic endometrial group,EMs The total methylation rate of the ectopic endometrium group and the methylation rate of infertile patients were significantly higher than those of the normal endometrium group(P<0.05).Conclusion The HOXA10 gene has abnormal DNA methylation of HOXA10 gene in EMs.The DNA methylation of HOXA10 gene is significantly higher than that of non-EMs infertility patients.
