宫内感染所致新生鼠肺损伤中MMP-9及MMP-11的表达分析 被引量:7

摘要 目的宫内感染所导致的新生鼠肺损伤中基质金属蛋白酶-9(MMP-9)与基质金属蛋白酶-11(MMP-11)表达的影响。方法选取健康雌性SD孕鼠,随机分为脂多糖组及对照组。脂多糖组分别在怀孕第19天腹腔注射0.4 mg/kg、0.6 mg/kg、0.8 mg/kg、1.0 mg/kg、1.2 mg/kg的脂多糖溶液,对照组在怀孕第19天注射等量生理盐水溶液。观察并记录不同组别孕鼠死亡率、新生鼠死亡率,HE染色观察肺部的炎症情况,并采用酶联免疫吸附试验检测新生鼠肺叶中MMP-9、MMP-11及炎症因子肿瘤坏死因子α(TNF-α)、白细胞介素-1(IL-1)、白细胞介素-6(IL-6)的表达情况。结果与对照组相比,脂多糖组新生鼠死亡率较高,差异具有统计学意义(P<0.05)。随着脂多糖浓度的上升,新生鼠的死亡率逐渐增高,且各组之间差异具有统计学意义(P<0.05)。脂多糖组新生鼠肺组织中MMP-9、MMP-11、TNF-α、IL-1、IL-6的浓度均高于对照组,具有统计学差异(P<0.05),脂多糖组随着脂多糖浓度的增加,较高浓度的脂多糖组新生鼠肺部MMP-9、MMP-11、TNF-α、IL-1、IL-6的表达均高于上一浓度梯度新生鼠的表达,差异具有统计学意义(P<0.05)。结论通过腹腔注射脂多糖能够成功建立宫内感染模型,宫内感染能够导致新生鼠死亡率上升及肺组织损伤,脂多糖组新生鼠肺中MMP-9、MMP-11及炎症因子的表达水平升高,且呈剂量依赖性。 Objective The effect of MMP-9(matrix metalloproteinase-9)expression in neonatal rat lung injury induced by intrauterine infection.Methods Healthy female SD rats were selected and randomly divided into a lipopolysaccharide group and a control group.The lipopolysaccharide group was intraperitoneally injected with 0.4 mg/kg,0.6 mg/kg,0.8 mg/kg,1.0 mg/kg,1.2 mg/kg lipopolysaccharide solution on the 19th day of pregnancy,and the control group was injected with the same amount of physiological saline solution on the 19th day of pregnancy.The mortality and neonatal mortality of different groups were observed and recorded.HE staining was used to observe the inflammation of the lungs.The expression of MMP-9,MMP-11,TNF-α,IL-1 and IL-6 in the lungs of neonatal rats was detected by enzyme-linked immunosorbent assay.Results Compared with the control group,the mortality of neonatal rats in the lipopolysaccharide group was higher,and the difference was statistically significant(P<0.05).With the increase of lipopolysaccharide concentration,the mortality of newborn rats gradually increased,and the difference between the groups was statistically significant(P<0.05).The concentrations of MMP-9,MMP-11,TNF-α,IL-1 and IL-6 in the lung tissue of the lipopolysaccharide group were higher than those in the control group(P<0.05).The expression of MMP-9,MMP-11,TNF-α,IL-1 and IL-6 in the lungs of the higher concentration of lipopolysaccharide group was higher than that of the newborn rats in the previous concentration gradient.The difference was statistically significant(P<0.05).Conclusion Intrauterine infection model can be successfully established by intraperitoneal injection of lipopolysaccharide.Intrauterine infection can lead to increased mortality and lung tissue damage,and elevated expression of MMP-9,MMP-11,inflammatory factor are found in the lungs in newborn rats in lipopolysaccharide group.
