腺苷A2b受体在大鼠脑缺血再灌注后的表达特点 被引量:1

The Characteristic of Adenosine A2b Receptor Expression after Cerebral Ischemia-reperfusion in Rats

摘要 目的探讨缺血/再灌注(I/R)后脑内腺苷A2b受体(A2bR)表达水平和分布的特点。方法SD大鼠建立大脑中动脉缺血模型,免疫荧光法检测I/R后A2bR在脑内表达的部位及细胞类型,Western blot检测A2bR表达的相对水平。结果Western blot结果显示大鼠缺血侧皮质、纹状体及海马区域A2bR表达均较对侧(非缺血侧)显著增高(P<0.05)。免疫荧光结果显示I/R后腺苷A2bR在神经元、血管内皮细胞和星形胶质细胞表达显著激活。结论I/R可诱导腺苷A2bR在缺血侧皮质、纹状体及海马区域的神经元、血管内皮细胞和星形胶质细胞表达。 Aim To explore the characteristic of level and distribution of adenosine A2b receptor(A2bR)expression after cerebral ischemia reperfusion in rats.Methods The distribution and cell types of A2 bR expression were demonstrated by immunofluorescence technique after cerebral ischemia reperfusion in rats.The level of A2 bR expression was detected by Western blot assay.Results Western blot results showed that the level of A2bR expression was increased significantly in the ipsilateral cortex,striatum and hippocampus(P<0.05).Immunofluorescence results showed that ischemia reperfusion could induce the A2bR activation of neuron,endothelial and astrocyte cells.Conclusion Ischemia reperfusion can activate A2bR expression in ipsilateral cortex,striatum and hippocampus,which mostly distribute in neuron,endothelial and astrocyte cells.
