ERK1/2信号通路的活化与大鼠急性肝衰竭关系的实验研究 被引量:4

摘要 目的 探讨ERK1/2信号通路的活化与大鼠急性肝衰竭(acute liver failure,ALF)的关系。方法 采用CCl 4溶液灌胃方式将SPF级Wistar雄性大鼠12只诱导成ALF模型作为实验组,另选择SPF级Wistar雄性大鼠12只作为正常对照组。观察两组大鼠的一般情况,并于48 h采集大鼠尾静脉血检测ALT、AST和PTA表达水平,随后处死大鼠,取肝组织进行HE染色,Western blotting检测ERK1/2和pERK1/2的表达,qRT-PCR检测ERK1/2 mRNA水平,并对结果进行统计学分析。结果 实验组ALT、AST、PTA、pERK1/2和ERK1/2 mRNA与正常对照组比较,差异均有统计学意义( P <0.01),但ERK1/2蛋白的表达差异无统计学意义( P > 0.05 )。结论 ALF的发生与ERK1/2信号通路的活化密切相关,推测ERK1/2信号通路的激活可能参与了ALF的发生机制。 Objective To investigate the relationship between the activation of ERK1/2 signaling pathway and acute liver failure(ALF) in rats.Methods Twelve SPF Wistar male rats were induced into ALF model by intragastric administration of CCl4 solution as experimental group. Another 12 SPF Wistar male rats were selected as normal control group. In the experiment, the general situation of rats in two groups was observed, the tail vein blood of rats was collected at 48 hours, and the expressions of ALT, AST and PTA were detected. Then the rats were sacrificed and the liver tissues were stained with HE. The expressions of ERK1/2 and pERK1/2 were detected by Western blotting and ERK1/2 mRNA was detected by qRT-PCR. The results were analyzed statistically.Results Expressions of ALT, AST, PTA, pERK1/2 and ERK1/2 mRNA in experimental group were significantly different compared with normal control group(P<0.01), but the expression of ERK1/2 protein was not significantly different(P>0.05).Conclusion The occurrence of ALF is closely related to the activation of ERK1/2 signaling pathway. It is speculated that the activation of ERK1/2 signaling pathway may be involved in the mechanism of ALF.
