摘要
目的研究DNATyper^TM19试剂盒的18个STR基因座在欧洲、非洲和东亚人群中的适用性。方法使用DNATyper^TM19复合扩增试剂盒,对欧洲人199份、非洲人188份和东亚人176份无关个体的DNA样本进行扩增,扩增产物用遗传分析仪3130xL进行检测,GenemapperIDv3.2软件对分型结果进行分析。结果在欧洲人中共检测出197种等位基因,等位基因频率分布在0.003~0.568之间;非洲人中共检测出193种等位基因,等位基因频率分布在0.003~0.436之间;东亚人中共检测出187种等位基因,等位基因频率分布在0.003~0.540之间;欧洲人、非洲人与东亚人的累积个体识别能力均大于99.9999999999%。结论DNATyper^TM19复合扩增试剂盒的18个STR基因座在欧洲、非洲、东亚人群中均具有较好的遗传多态性,对三大人群均具有较好的适用性。
Objective To investigate the applicability of DNA Typer^TM 19 kit to distinguish European, African and East Asian population. Methods 563 unrelated individuals’ DNA samples, among which 199 pieces are of European, 188 of African and 176 of East Asian, were co-ampli ed with DNA Typer^TM 19 kit to have their 18 STR loci cloned. The ampli ed PCR products were capillary-electrophoresed through 3130XL genetic analyzer, together with their genotyping results being analyzed by Genemapper ID v3.2. Results 197 alleles were identi ed from the European, with their allelic frequencies ranging from 0.003-0.568;193 alleles were found from the African, unveiling their allelic frequencies spanning 0.003-0.436;187 alleles were discovered from the East Asian, bestriding their allelic frequencies over 0.003-0.540. The TDP was greater than 99.999 999 999 9% for all the three populations of European, African and East Asian. Conclusions The DNA Typer^TM 19 kit can be used to analyze genetic polymorphism into European, African and East Asian populations.
