摘要 目的:探讨人脐带间充质干细胞(hUCMSCs)对早发性卵巢功能不全模型大鼠卵巢功能的影响.方法:将正常动情周期的36只雌性ICR小鼠按随机数字表法分为移植组(n=12)、模型对照组(n=12)与空白对照组(n=12).空白对照组大鼠不进行任何处理,模型对照组和移植组大鼠腹腔注射顺铂(CDDP)建立早发性卵巢功能不全(POI)模型.移植组通过尾静脉注射hUCMSCs进行治疗,模型对照组与空白对照组均注射等量灭菌用水.移植后2周,观察三组一般情况,HE染色三组的切片中各阶段的卵泡情况,同时观察卵巢结构变化.测定血清中雌二醇(E2)、卵泡刺激素(FSH)、黄体生成素(LH)和抗苗勒管激素(AMH)水平,并分析卵巢储备功能的变化.结果:电镜下显示,模型对照组颗粒细胞核溶解、核膜消失、线粒体空泡化,线粒体与内质网数量逐渐减少.移植组颗粒细胞细胞核膜逐渐修复,核仁出现,内质网与线粒体少量存在.造模后3 d,模型对照组E2含量低于空白对照组,FSH含量高于空白对照组(P<0.05).移植后2周,移植组AMH含量高于模型对照组(P<0.05),三组AMH比较,差异有统计学意义(P<0.05).结论:hUCMSCs移植对POI大鼠的卵巢结构和功能具有一定的修复作用. Objective:To investigate the effect of human umbilical cord mesenchymal stem cells (hUCMSCs)on ovarian function in rats with early-onset ovarian dysfunction.Method:36 female ICR mice with normal estrous cycle were divided into transplantation group(n=12),model control group(n=12)and blank control group(n=12)according to random number table method.The blank control group without any treatment. The model control group and transplantation group rats were intraperitoneally injected with cisplatin(CDDP)to establish premature ovarian insufficiency(POI)model.The transplantation group was treated with intravenous injection of hUCMSCs.The model control group and blank control group were injected with the same amount of sterilizing water.2 weeks after transplantation,the general conditions of the three groups were observed,and the follicular conditions at each stage in the sections of the three groups with HE staining were observed,and the structural changes of the ovary were observed at the same time.The levels of estradiol(E2),follicle stimulating hormone(FSH),luteinizing hormone(LH)and anti-Mullerian hormone(AMH)in serum were measured, and the changes of ovarian reserve function were analyzed.Result:Electron microscopy showed that the number of mitochondria and endoplasmic reticulum decreased gradually in the model control group,while the number of nuclear membrane repaired gradually,nucleolus appeared,and endoplasmic reticulum and mitochondria existed little in the transplantation group.3 days after modeling,E2 content in the model control group was lower than that in the blank control group,and FSH content was higher than that in the blank control group(P<0.05).At 2 weeks after transplantation,AMH content in the transplantation group was higher than that in the model control group (P<0.05).AMH in the three groups were compared,the differences were statistically significant(P<0.05). Conclusion:hUCMSCs transplantation can repair the ovarian structure and function of POI rats.
