摘要
目的:探讨中医方剂治疗胃癌的核心药物及其干预靶点。方法:通过复杂网络的方法基于近20年文献筛选中药方剂中治疗胃癌的主药,再通过中药网络药理学数据库以及TCGA肿瘤全基因组数据库,采用数据挖掘方法探索预测主药治疗胃癌的机制及关键靶点,并通过大数据高通量全基因组信息观察调节关键靶点对胃癌患者生存期的影响。结果:健脾燥湿补气类中药“白术-茯苓-黄芪”对CXCR4可能存在相关调节作用并对肿瘤患者生存周期具有显著影响,这可能是中医方剂治疗胃癌的新机制之一,并可结合方药的核心处方和干预靶点优化方剂配伍,这对既往“以证测方,以症拟方”的思路是一种新的补充。结论:“白术-茯苓-黄芪”共同调节的潜在基因靶点是CXCR4,这可能是“白术-茯苓-黄芪”的起效机制之一。
Objective: To explore the core drugs of traditional Chinese medicine (TCM) in the treatment of gastric cancer and their intervention targets. Methods: The main drugs for gastric cancer in traditional Chinese medicine prescriptions were screened based on the literature of the past 20 years by the complex network method. Based on the network pharmacology database of traditional Chinese medicine and TCGA tumor genome database, the data mining method was used to explore the mechanism and key targets for predicting the treatment of gastric cancer by main drugs. Then the effects of key regulatory targets on the survival of gastric cancer patients were observed with large data and high -throughput genome -wide information. Results: Chinese herbal Baizhu (Atractylodes macrocephala)-Fuling (Poria cocos)-Huangqi (Astragalus membranaceus) may have a related regulatory effect on CXCR4 and have a significant effect on the survival cycle of tumor patients. This may be one of the new mechanisms for the treatment of gastric cancer with traditional Chinese medicine. The core prescription and intervention target optimization prescription compatibility, which is a new supplement to the previous idea of "testing the party and symptomatic". Conclusion: The potential gene target for the joint regulation of Baizhu-Fuling-Huangqi is CXCR4, which may be one of the mechanisms of Baizhu-Fuling-Huangqi.
