重楼皂苷Ⅱ对高糖干预下肾小球系膜细胞增殖及氧化应激的影响 被引量:4

摘要 目的探讨重楼皂苷Ⅱ对高糖干预下肾小球系膜细胞(GMC)增殖及氧化应激损伤的影响。方法将GMC分为正常对照组、甘露醇对照组、高糖组和重楼皂苷Ⅱ组,分别用含有5.6 mmol/L葡萄糖、5.6 mmol/L葡萄糖+19.5 mmol/L甘露醇、25 mmol/L葡萄糖、25 mmol/L葡萄糖+20μg/mL重楼皂苷Ⅱ的杜氏改良伊格尔培养基培养24 h。检测4组细胞的增殖活性、丙二醛含量、超氧化物歧化酶(SOD)活性以及活性氧含量。结果与正常对照组比较,高糖组、重楼皂苷Ⅱ组细胞的增殖活性、丙二醛含量、活性氧含量升高,SOD活性降低(P<0.05);与高糖组比较,重楼皂苷Ⅱ组细胞的增殖活性、丙二醛含量、活性氧含量减低,SOD活性升高(P<0.05)。结论重楼皂苷Ⅱ对高糖干预下GMC增殖有一定的抑制作用,并可减轻细胞氧化应激损伤,这可能与其减少细胞内活性氧产生和提高SOD活性有关。 Objective To investigate the effects of Polyphyllin Ⅱ on proliferation and oxidative stress injury in glomerular mesangial cell(GMC) under high glucose intervention. Methods GMCs were divided into normal control group, mannitol control group,high glucose group and Polyphyllin Ⅱ group,cultured for 24 hours in Dulbecco′s modified Eagle medium containing 5.6 mmol/L glucose,5.6 mmol/L glucose+19.5 mmol/L mannitol, 25 mmol/L glucose, 25 mmol/L glucose+20 μg/mL Polyphyllin Ⅱ,respectively.Proliferation activity,content of malondialdehyde, activity of superoxide dismutase(SOD),and content of reactive oxygen species were detected in the cells of the four groups. Results Compared with the normal control group,the high glucose group and the Polyphyllin Ⅱ group had increased proliferation activity,elevated contents of malondialdehyde and reactive oxygen species,and decreased SOD activity ( P <0.05);compared with the high glucose group,the Polyphyllin Ⅱ group exhibited lower proliferation activity,and decreased contents of malondialdehyde and reactive oxygen species,as well as elevated SOD activity( P <0.05). Conclusion Polyphyllin Ⅱ has a certain inhibitory effects on GMC proliferation under high glucose intervention,and can attenuate oxidative stress injury,which might correlate with its role in reducing the production of reactive oxygen species in the cells and elevating SOD activity.
