摘要
目的:研究绞股蓝总皂苷对动脉粥样硬化(AS)小鼠血管PPAR-γ/LXR-α/ABCA1信号通路的影响.方法:高脂饲养ApoE-/-小鼠建立动脉粥样硬化模型动物,并用200 mg·kg-1和400 mg·kg-1剂量的绞股蓝总皂苷进行干预,化学法检测小鼠血脂水平,HE染色法检测主动脉组织病理学变化,荧光定量PCR和免疫印迹技术检测血管中PPAR-γ、LXR-α及ABCA1的表达.结果:与正常组相比,AS模型组小鼠血清中TC、TG、LDL-C含量显著升高(P<0.05),血管内动脉粥样硬化斑块面积增加.绞股蓝总皂苷干预后,TC、TG、LDL-C含量明显降低(P<0.05),AS斑块减小,PPAR-γ、LXR-α、ABCA1表达显著增加(P<0.05).结论:绞股蓝总皂苷能够降低AS小鼠血脂含量,并可能通过激活PPAR-γ/LXR-α/ABCA1信号通路从而发挥抗动脉粥样硬化的作用.
[Objective]To study the effect of gypenosides on PPAR -γ/LXR -α/ABCA1 signaling pathway in atherosclerosis mice vascular.[Method]The model of atherosclerosis was established by ApoE^-/- mice were fed with high-fat diet, and treated with 200 mg·kg^-1 and 400 mg·kg^-1 Gypenosides. The blood lipid levels were measured by chemical methods and detection aortic tissue pathology by HE staining. The expression of PPAR -γ,LXR -α and ABCA1 and ABCA1 in vascular were detected by fluorescence quantitative PCR and immunoblotting.[Result]Compared with the normal group, the levels of serum TC, TG and LDL-C in AS model group were significantly increased (P < 0.05 ), and the area of intravascular plaque increased. After treated with Gypenosides, the levels of TC, TG LDL-C and plaques area were significantly decreased (P < 0.05 ), and the expression of PPAR-γ, LXR-α and ABCA1 was significantly increased (P < 0.05 ).[Conclusion]Gypenosides can reduce the lipid levels of AS mice, and may play a role of anti-atherosclerosis by activating PPAR -γ/LXR-α/ABCA1 signaling pathway.
