摘要
以嗜酸乳杆菌102为出发菌株,通过单因素试验探究嗜酸乳杆菌102的最适生长条件为:培养温度36℃、pH6.0、接种量3%,并绘制出该菌种的生长曲线。在此条件下进行以玉米淀粉糖副产物-玉米浆和结晶葡萄糖母液为主要原料,发酵培养基的单因素初选试验、响应面优化试验,结果表明,两两因素间存在比较明显的交互作用,最佳点落在试验考察区域内。通过软件分析优化后的发酵培养基配方为玉米浆8.699%,结晶糖母液5.360%,低聚异麦芽糖0.669%。利用3L全自动发酵罐,在最适培养条件下培养15h,最终发酵液中活菌数达到14.960×10^9CFU/ml,与模型拟合度为97.75%,可靠性高,为淀粉糖副产物综合利用拓宽了思路。
In order to increase the added value of corn starch sugar by-products and reduce the production cost of Lactobacillus acidophilus,Lactobacillus acidophilus 102 was chosed as the starting strain,the optimum growth conditions of Lactobacillus acidophilus 102 were studied by single factor experiment:culture temperature 36℃,pH 6.0,inoculation amount 3%,and draw the growth curves of the bacteria.Under these conditions,the single factor experiment and response surface optimization experiment were carried out on the fermentation medium which was mainly made of corn pulp and crystalline glucose mother liquor.The results showed that there was obvious interaction between the two factors.The optimum point was within the experimental investigation area.Through the analysis of software,the optimized fermentation medium formula was as follows: Corn Steep Liquor 8.699%,Crystalline glucose mother liquor 5.360%,Isomaltooligosaccharide 0.669%.Using 3L automatic fermentor,culture for 15 hours under the optimum conditions,the number of viable bacteria in the final fermentation broth reached 14.960×10^9 CFU/ml,The fitting degree with the model is 97.75%.Fermentation of Lactobacillus acidophilus using corn starch sugar by product Corn Steep Liquor and crystalline glucose mother liquor as main raw materials is feasible.This study broadened the thinking for the comprehensive utilization of starch sugar by products.
