铁皮石斛纤维素合成酶超级基因家族生物信息学分析 被引量:7

摘要 纤维素合成酶超级基因家族是植物体纤维素合成的重要基因。为研究铁皮石斛纤维素合成酶和类纤维素合成酶基因(CESA/CSL)家族的功能,利用从铁皮石斛转录组中鉴定出的32个在茎中高表达量的纤维素合成酶超级基因家族成员为研究对象,使用MEGA5.0、GSDS、SMART、TMHMM、WoLFPSORT等软件对其构建系统进化树,并对基因结构及蛋白质保守结构域、跨膜结构和亚细胞定位等进行综合分析。结果显示,CESA/CSL基因在铁皮石斛不同组织中表达的数目和表达量均不同,具有组织表达特异性。铁皮石斛茎中表达量较高的CESA/CSL基因家族可分为6个亚族,外显子数目1~21个。CESA/CSL基因编码的蛋白质保守性较强,除CSLA亚族外,其余亚族均含有该基因家族的保守结构域Cellulosesynthase。CESA/CSL蛋白主要分布于质膜上,大部分具有跨膜结构域。 Cellulose synthase supergene family plays an important role in cellulose synthesis of plants.In order to study the function of CESA/CSL gene family of Dendrobium candidum ,taking 32 members of cellulose synthase supergene family identified from the transcription group of Dendrobium candidum with high expression in stem as the objects and using MEGA 5.0,GSDS,SMART,TMHMM,WoLF PSORT and other softwares,the phylogenetic tree was established and the gene structure,protein conserved domain,transmembrane structure and subcellular localization were analyzed comprehensively.The results showed that the CESA/CSL gene in different tissues of Dendrobium candidum was expressed differently in number and expression level,and had the specificity of tissue expression.The CESA/CSL gene family with high expression in the stem of Dendrobium candidum could be divided into 6 subgroups,and the number of exons was 1-21.The proteins encoded by CESA/CSL gene family were highly conserved,and contained the conserved domain Cellulose synthase except for the CSLA subfamily.In addition,CESA/CSL protein was mainly distributed on the plasma membrane,and most of them had transmembrane domains.
