引发剂对黄芩种子萌发及幼苗耐旱性的影响 被引量:2

摘要 为选择适合黄芩生长的引发剂,提高人工栽培黄芩的产量和质量,以商洛黄芩种子及1年生黄芩幼苗为材料,采用20%PEG-6000模拟干旱条件,设蒸馏水处理(CK1)和20%PEG-6000处理(CK2)2个对照,研究了3种单引发剂和8种复合引发剂对黄芩种子萌发和幼苗耐旱性的影响。结果表明,单引发剂处理黄芩种子以20mmol/LCaCl2效果最好,黄芩种子发芽势和发芽率较CK2分别增加120.47%和103.66%;复合引发剂以0.3%NaCl+5mmol/LNa2SO3+15mmol/LCaCl2处理效果最佳,黄芩种子发芽势和发芽率较CK2分别增加97.91%和86.94%;2种引发剂比较以20mmol/LCaCl2效果最好。用单引发剂处理黄芩幼苗以0.4%NaCl效果最佳,叶绿素、可溶性蛋白、脯氨酸含量分别比CK2增加147.75%、182.47%、166.47%,超氧化物歧化酶(SOD)、过氧化物酶(POD)、过氧化氢酶(CAT)活性较CK2分别增加41.75%、80.29%、82.24%,MDA含量较CK2降低65.94%;复合引发剂处理黄芩幼苗以0.4%NaCl+5mmol/LNa2SO3+15mmol/LCaCl2效果最佳,叶绿素、可溶性蛋白、脯氨酸含量较CK2分别增加114.61%、146.75%、146.83%;SOD、POD、CAT活性较CK2分别增加34.42%、65.69%、64.80%;2种引发剂相比以0.4%NaCl效果最佳。可见,种子引发能有效提高黄芩种子的萌发率及幼苗的耐旱性,单引发剂处理种子以20mmol/LCaCl2效果最好,复合引发剂处理种子以0.3%NaCl+5mmol/LNa2SO3+15mmol/LCaCl2效果最佳;单引发剂处理黄芩幼苗以0.4%NaCl效果最佳,复合引发剂处理黄芩幼苗以0.4%NaCl+5mmol/LNa2SO3+15mmol/LCaCl2效果最佳。 In order to select the suitable initiator for the growth of Scutellaria baicalensis ,and improve the yield and quality of artificially cultivated Scutellaria baicalensis ,the seeds and annual seedlings of Shangluo Scutellaria baicalensis were used as materials,using 20% PEG-6000 to simulate drought conditions,and distilled water treatment(CK 1) and 20% PEG-6000 treatment(CK2) were used to study the effects of three single initiators and eight complex initiators on the seed germination and drought tolerance of Scutellaria baicalensis.The results showed that 20 mmol/L CaCl2 was the best single initiator for treating Scutellaria baicalensis seeds.Compared with CK 2,the germination potential and germination rate of Scutellaria baicalensis seeds increased by 120.47% and 103.66% respectively.The best treatment of composite initiator was 0.3% NaCl+5 mmol/L Na 2SO 3+15 mmol/L CaCl2.Compared with CK 2,the germination potential and germination rate of Scutellaria baicalensis increased by 97.91% and 86.94% respectively.The comparison of the two initiators showed that 20 mmol/L CaCl2 was the best.The best effect of treating Scutellaria baicalensis seedlings with single initiator was obtained by 0.4% NaCl.Compared with CK 2,chlorophyll,soluble protein and proline contents increased by 147.75%,182.47% and 166.47% respectively,the activities of superoxide dismutase(SOD),peroxidase(POD) and catalase(CAT) increased by 41.75%,80.29% and 82.24% respectively,MDA content reduced by 65.94%.The best effect of treating Scutellaria baicalensis seedlings with complex initiator was obtained by 0.4% NaCl+5 mmol/L Na 2SO 3 +15 mmol/L CaCl 2.Compared with CK2,chlorophyll,soluble protein and proline contents increased by 114.61%,146.75% and 146.83% respectively,the activities of SOD,POD and CAT increased by 34.42%,65.69% and 64.80% respectively.By comparing the two initiators,the best initiator was 0.4% NaCl.It can be seen that seed initiation can effectively improve the germination rate of Sctellaria baicalensis seeds and the drought tolerance of seedlings.The effect oF20 mmol/L CaCl2 is the best for treating seeds by single initiator,0.3% NaCl + 5 mmol/L Na 2SO 3+15 mmol/L CaCl2 is the best for treating seeds by composite initiator,0.4% NaCl is the best for treating Scutellaria baicalensis seedlings by single initiator,and 0.4% NaCl+5 mmol/L Na2SO3+15 mmol/L CaCl2 is the best for treating Scutellaria baicalensis seedlings by composite initiator.
