大脑类器官模型在精神疾病研究中的现状及展望

摘要 精神疾病是常见的病因复杂、发病率高、多基因遗传疾病,造成了极大的经济负担和社会负担。目前各种体内外模型已广泛用于研究精神疾病的病因学机制和精神药物相关研究。然而,由于精神疾病的复杂性,如何选用合适的模型从结构、细胞和蛋白水平更好地研究精神障碍的发病机制和药物作用,具有重要的临床意义[1]。大脑类器官(cerebral organoids)是由干细胞定向诱导分化形成的大脑皮质结构,可一定程度上在体外模拟人类胚胎早期大脑的发育过程和结构特点,在研究精神疾病的起源和病理学、药物筛选和基因改造方面显示了很大潜力[2]。本综述对大脑类器官在精神医学的研究进展与局限性进行探讨,并对大脑类器官未来的发展和应用进行评估与展望。
