白芨试管苗高效增殖方法试验研究 被引量:2

摘要 以白芨(Bletilla striata(Thunb.)Reichb.f.)成熟未开裂的蒴果的种子为外植体,将白芨种子播种在含有不同浓度生长激素的培养基上,待种子萌发后,利用组织培养技术进行无性繁殖,研究不同生长激素浓度对种子萌发率、丛生芽增殖率、生根培养的影响,并进行炼苗移栽,建立了高效增殖体系.试验结果表明:1/2MS+1.0mg/L1-萘乙酸(1-Naphthaleneacetic acid,NAA)培养基最适于种子萌发,丛生芽在MS+0.5mg/LNAA+1.0mg/L6-苄氨基腺嘌呤(6-Benzylaminopurine,6-BA)培养基上增殖效果最好,1/2MS+0.5mg/LNAA培养基生根效果最好.炼苗基质为泥炭土:蛭石:珍珠岩(3∶1∶1),成活率可达90%以上.最适宜的炼苗温度是23℃~28℃,空气相对湿度是80%~90%时有利于提高炼苗成活率. Using mature uncracked capsule seeds of Bletilla striata (Thunb.)Reichb.f.as explants, seed germination, adventitious bud induction, proliferation and rooting rates were studied to see the effects of growth hormones, and to select suitable combinations.The seedlings from tissue culture were then transplanted.The results showed that the medium of1/2MS +1.0m g/L NAA was the most suitable for the seed germination, and the regeneration buds had the highest proliferation rate on medium of MS +0.5mg/L NAA +1.0 mg/L6-BA.1/2MS + 0.5m g/L NAA was ideal for the growth of roots.We transplanted them on the substrate composed of3 parts of peaty soil and1 part of vermiculite and1 part of pearlstone.The survival rate was more than 90 %.The seedling survival rate could be improved when the temperature was25℃-28℃ and air relative humidity was 80%-90%.
