氧化苦参碱抗大鼠胰腺星状细胞纤维化作用机制研究 被引量:5

摘要 目的研究氧化苦参碱(OM)对大鼠胰腺星状细胞(PSCs)(LTC14细胞株)纤维化的作用机制。方法取LTC14细胞,分别设为对照组、模型组、实验组,对照组进行常规培养,模型组使用转化生长因子β1(TGF-β1)处理,实验组采用TGF-β1+OM处理。作用12h后收集细胞培养基上清,提取蛋白及RNA;检测并比较细胞中α-平滑肌肌动蛋白(α-SMA)、纤连蛋白(FN)、I型胶原(CoL-I)、NOD样受体蛋白3(NLRP3)、半胱氨酸天冬氨酸蛋白酶(caspase1)、白介素1β(IL-1β等)指标的蛋白和RNA表达量。结果与对照组相比,模型组α-SMA、FN、CoL-I、NLRP3、caspase1、IL-1β蛋白及mRNA表达均呈显著增高趋势(P<0.01);与模型组比较,实验组α-SMA、FN、CoL-I、NLRP3、caspase1、IL-1β蛋白及mRNA表达均显著降低(P<0.01)。结论OM可能通过抑制NLRP3炎性小体活化而产生抗纤维化作用。 Objective To study action mechanism of Oxymatrine(OM)on fibrosis of pancreatic stellate cells(PSCs,LTC14 cell line)in rats.Methods LTC14 cells were divided into control group,model group and experimental group.Control group was cultured routinely,while model group was treated with transforming growth factorβ1(TGF-β1)),and experimental group was treated with TGF-β1+OM.After 12 h of treatment,supernatant samples of cell culture medium were collected,and protein and RNA samples were got out.Protein and RNA expressions ofα-smooth muscle actin(α-SMA),fibronectin(FN),type I collagen(CoL-I),NOD-like receptor protein 3(NLRP3),caspase 1(caspase 1),interleukin 1β(IL-1β)were detected and compared.Results Compared with those in control group,protein and RNA expressions ofα-SMA,FN,CoL-I,NLRP3,caspase-1 and IL-1βin model group were significantly increased(P<0.01).Compared with those in model group,protein and RNA expressions ofα-SMA,FN,CoL-I,NLRP3,caspase-1 and IL-1βin experimental group were significantly decreased(P<0.01).Conclusion OM may have anti-fibrosis effect by inhibiting activation of NLRP3 inflammatory bodies.
